  • General notes

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Target

  • Function

Apoptotic suppressor. The BIR motifs region interacts with TNF receptor associated factors 1 and 2 (TRAF1 and TRAF2) to form an heteromeric complex, which is then recruited to the tumor necrosis factor receptor 2 (TNFR2).

Present in many fetal and adult tissues. Mainly expressed in adult skeletal muscle, thymus, testis, ovary, and pancreas, low or absent in brain and peripheral blood leukocytes.
  • Sequence similarities

    Belongs to the IAP family.
    Contains 3 BIR repeats.
    Contains 1 CARD domain.
    Contains 1 RING-type zinc finger.

      Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-cIAP1 antibody (ab2399)
IHC image of ab2399 staining in human tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab2399, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    Question

the monoclonal antibody we are using is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Leu28 of human c-IAP1 protein.

The polyclonalantibody we are usingis produced by immunizing animals with a synthetic peptide corresponding to a region between the first two BIR domains of c-IAP1.

    Thank you for your help.

    Answer

    Thank you for your email.

The peptide is from the sequence around amino acid 530 of human cIAP1. This sequence does not match with the immunogen sequence of polyclonal and monoclonal antibodies you have so this peptide ab38272 would be of no use.

If you are interested in doing blocking experiment then ab38272 would block ab2399.

I hope this information will be helpful. Should you have any question please do not hesitate to contact me.

    Question

I would like to test the specificity ofone of our anti-cIAP1 mAb by using your cIAP1 blocking peptide.

I would therefore know if this peptide can be used as an immunogen for other antibodies than https://www.abcam.com/cIAP1-antibody-ab2399.html.

Thank you in advance for your kind reply.

    Asked on Sep 14 2012

    Answer

    Thank you for contacting us.

Could you please explain what do you mean by other antibodies? The antibodies against the same peptide should be same however difference can be seen in specificity, purity etc.

The peptide ab38272 is from a sequence flanking 530 of human cIAP1. We unfortunately don't know the exact sequence so this peptide may not able to block your antibody.

Do you know the immunogen sequence of your antibody?

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Thank you for your inquiry. We source this antibody from an outside laboratory, and unfortunately we do not have access to the exact sequence used for the immunogen. If you plan to use the antibody on a fragment of the protein, we may be able to get more specific information of whether this antibody would bind to your fragment, but unfortunately we don't have access to any further information than what is on the datasheet.
