Target

• Function

Histone H3-like variant which exclusively replaces conventional H3 in the nucleosome core of centromeric chromatin at the inner plate of the kinetochore. Required for recruitment and assembly of kinetochore proteins, mitotic progression and chromosome segregation. May serve as an epigenetic mark that propagates centromere identity through replication and cell division.
• Sequence similarities

Belongs to the histone H3 family.
• Cellular localization

Nucleus. Chromosome, centromere, kinetochore. Localizes exclusively in the kinetochore domain of centromeres. Co-expressed with yemalpha.
• Target information above from: UniProt accession Q9V6Q2 The UniProt Consortium
The Universal Protein Resource (UniProt) in 2010
Nucleic Acids Res. 38:D142-D148 (2010) .

Question

We unfortunately were not able to obtain a better result by changing our
fixation protocol. Therefore, we ask that you issue us a credit against
our purchase of a replacement vial of ab10887 from the new batch that has
just gone into production. We understand that the new batch is still
several weeks away from being ready for sale.
Can you enter an order for us now, to be shipped when the new batch of
ab10887 actually is ready to go on sale? If not, please let me know what
is the best way for us to obtain a vial from the new batch.

Answer

Thank you for your reply. Unfortunately, since the new lot is still in production, I am unable to set up an automatic alert for when it comes into stock or an order for a lot that doesn't exist yet. I have issued you a refund (Credit Note ***) and will check periodically for the new lot of ab10887. Once the new batch is available, you can place the order like normal and specify the lot number I will send you. The charge for the antibody and shipping / packing will be refunded automatically to the card used to make the purchase. If you have any questions about this refund, please contact us.credits@abcam.com.

Question

Thank you for looking into this. Given the time frame involved, it
doesn't seem like we will be able to defer our work until the next batch
of ab10887 becomes available.
Before we request a refund, however, we would like to see whether the
antibody might perform any better for us if we use a methanol fixation
protocol. We can do the experiment either this Friday or next Monday, and
if we continue to get high background we would then get back to you right
away in order to seek a refund.
So, I guess I need to know whether you are able to wait a few more days
for us to make a decision?

Asked on Jul 10 2012

Answer

Thank you for your patience. I checked with my colleagues in our production lab and we are currently scheduled to begin immunizations for a new batch of ab10887 in one week. This would mean that a new lot would be available for sale in a couple of months. Unfortunately, I am unable to give a more accurate timeframe because it will depend on how the antibody performs in QC.

We do currently have additional stock of the same batch of ab10887 and I would be happy to send you a new vial if you would like to try one. Alternatively, I could offer you a replacement with an antibody to a different target, or a credit or refund. Please let me know how you would like to proceed and I will be happy to assist you further.

Asked on Sep 06 2011

Answer

According to our records, ab10887 was showing lot-to-lot variation and we were in contact in order to help resolve the issue. Looking at our correspondence, it appears that we are awaiting some images to forward to the lab, and we would be happy to send a new lot if we have one in stock. If the issue has already been settled, please let us know so that we can be assured that the problem has been solved to your satisfaction and update our records. We wish you the best of luck with your research and look forward to hearing from you. Please let us know if there is anything else that we can do for you.
