Target

  • Function
    Claudins function as major constituents of the tight junction complexes that regulate the permeability of epithelia. While some claudin family members play essential roles in the formation of impermeable barriers, others mediate the permeability to ions and small molecules. Often, several claudin family members are coexpressed and interact with each other, and this determines the overall permeability. CLDN1 is required to prevent the paracellular diffusion of small molecules through tight junctions in the epidermis and is required for the normal barrier function of the skin. Required for normal water homeostasis and to prevent excessive water loss through the skin, probably via an indirect effect on the expression levels of other proteins, since CLDN1 itself seems to be dispensable for water barrier formation in keratinocyte tight junctions (PubMed:23407391).
    (Microbial infection) Acts as a receptor for hepatitis C virus in hepatocytes (PubMed:17325668). Acts as a receptor for dengue virus (PubMed:24074594).

3. How long were the cells fixed for?

4. You have mentioned you also wanted to use the antibody in Western blotting, have you used ab15099 in this application?

5. What blocking conditions are you using?

6. Could you provide the order number for ab15099? Or if you do not have this, the delivery address used?

Once I have received this information I will look at the protocol and see if there are any suggestions we can make that may improve the results.

Having reviewed this case, I would like to offer some suggestions to help optimize the results from ab15099. I would also appreciate if you can confirm some further details:
Are your rat intestinal tissues paraffin-embedded (IHC-P) or frozen (IHC-Fr)? I was confused by the multiple fixatives used so I wasn't sure which technique was tried.
If you're using PFA, you will have to try a permeabilization step as the epitope may be intracellular since the immunogen is C-terminal. Use 0.1 to 0.2% Triton-X 100 in PBS for 10 min only. You shouldn't need it for acetone fixation.
You hadn't tried the recommended antigen retrieval as stated on the datasheet. It says, "Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Citrate buffer, pH 6.0 for 10 min followed by cooling at RT for 20 min."
You may want to try blocking in 10% serum from the species that your secondary antibody is raised in instead of 5% BSA.
Also, this antibody is prediluted, meaning that it should be used neat (or right out of the vial). Do not dilute it at all, but just put it on the slide. This may improve the signal.
What dilution of the secondary was used? You may want to increase the amount used.
Should the suggestions not improve the results, please do let me know.
