WB Use a concentration of 2 µg/ml. Detects a band of approximately 30 kDa (predicted molecular weight: 25 kDa). Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.

  • Relevance

    Claudin proteins are a family of transmembrane proteins associated with tight junctions, and different tissues exhibit different Claudin composition. There are 18 homolgs present in humans, and mutations in any of these proteins cause a variety of human disease, this indicates their physiological importance. CLDN17, clustered with CLDN8 at human chromosome 21q22.11, is a four transmembrane protein with WWCC motif, defined by W-X(17-22)-W-X(2)-C-X(8-10)-C.

Based on the information in your review, I feel that altering the following steps in your protocol may help improve your results:
- blocking less long (e.g. 1h)
- trying a dfferent fixation method (e.g. PFA, acetone)
- using more primary antibody (1/100)
- incubating longer with the primary antibody (overnight 4 degree)
- checking that the secondary antibody is working well with other primaries.

Thank you for getting back to me. Generally if an immunogen forms an alignment of 6 or more consecutive residues I would consider it possible that the antibody would react against the homolog of the species. However, if it is a polyclonal antisera this is not always the case as given its polyclonal nature it is a mixture of immunoglobulin molecules. I would be very interested to know the results of your sequence comparisons as I could not find an immunogen that carried significant sequence similarity to warrant the suggestion of the application of the antibody to fish. I hope this information helps, please do not hesitate to contact us if you need any more advice or information.

Thank you for your enquiry. I have performed a sequence analysis of your fish sequence with all of immunogens used to raise our antibodies against Claudin. Unfortunately none had significant sequence similarity that would make me consider reactivity likely. I am sorry that I cannot be of more assistance on this occasion.
