    Caspases are synthesized as inactive pro-enzymes that are processed to active form in cells undergoing apoptosis. Caspase 3 has been extensively studied and implicated to play an important role in apoptosis. Active caspase 3 proteolytically cleaves and activates other caspases, as well as relevant targets in the cells (e.g., PARP). This affinity purified antibody recognizing the active forms of caspase-3 provides a new tool for identifying apoptotic cell populations in both tissue sections and cultured cells.

Target

  • Function

    Involved in the activation cascade of caspases responsible for apoptosis execution. At the onset of apoptosis it proteolytically cleaves poly(ADP-ribose) polymerase (PARP) at a '216-Asp-
    -Gly-217' bond. Cleaves and activates sterol regulatory element binding proteins (SREBPs) between the basic helix-loop-helix leucine zipper domain and the membrane attachment domain. Cleaves and activates caspase-6, -7 and -9. Involved in the cleavage of huntingtin. Triggers cell adhesion in sympathetic neurons through RET cleavage.
  • Tissue specificity

    Highly expressed in lung, spleen, heart, liver and kidney. Moderate levels in brain and skeletal muscle, and low in testis. Also found in many cell lines, highest expression in cells of the immune system.
  • Sequence similarities

    Belongs to the peptidase C14A family.
  • Post-translational
    modifications

    Cleavage by granzyme B, caspase-6, caspase-8 and caspase-10 generates the two active subunits. Additional processing of the propeptides is likely due to the autocatalytic activity of the activated protease. Active heterodimers between the small subunit of caspase-7 protease and the large subunit of caspase-3 also occur and vice versa.
    S-nitrosylated on its catalytic site cysteine in unstimulated human cell lines and denitrosylated upon activation of the Fas apoptotic pathway, associated with an increase in intracellular caspase activity. Fas therefore activates caspase-3 not only by inducing the cleavage of the caspase zymogen to its active subunits, but also by stimulating the denitrosylation of its active site thiol.

    3) Description of the problem: We have been testing an antibody anti-active Caspase 3 that was purchased from Abcam (ab2302). When we tested the positive control that you suggest (Camptothecin 2 µM treated Jurkat cells) we obtained a very weak signal using the antibody at the concentration 1:100. We already tried several protein concentrations as well as different denaturation temperatures and transfer times. The results were always as you can see in the picture or even worst. Hence, we would like to know if you have any suggestion.

    Lysis buffer: RIPA
    Protease inhibitors: protease inhibitor cocktail provided with RIPA

    Thank you for taking time to complete our questionnaire. I am sorry to hear that this antibody is not providing satisfactory results.


    Having reviewed this case, I would like to offerthe followingsuggestions- all the other steps seem to be optimized already:

    1.) I can suggest to use BSA as a blocking agent. Indeed, changing the blocking agent can significantly improve the results. As an illustration for this effect, you can also see the WB image on the datasheet of the ab9385: https://www.abcam.com/HRP-GAPDH-antibody-Loading-Control-ab9385.html (or use the following: https://www.abcam.com/HRP-GAPDH-antibody-Loading-Control-ab9385.html).

    2.) What type of ECL are you using? I can recommend to use a ECL plus or supersensitive for the detection. (e.g. ab133409 https://www.abcam.com/ECL-Substrate-Kit-Ultra-High-Sensitivity-ab133409.html (or use the following: https://www.abcam.com/ECL-Substrate-Kit-Ultra-High-Sensitivity-ab133409.html)..) xx.

    I am sorry to hear you are experiencing difficulties with one of our products. We take product complaints very seriously, and investigate every product that we feel may not be performing correctly. I am not aware of any problems with this antibody.

    I would like however to investigate this in more details. Can you please send me the technical details of the protocol as well? Additionally, please send me the order number or the date on which the antibody has been ordered. This will allow me to identify the vial.

    I am attaching our questionnaire so that we can gather further information regarding the samples tested and the protocol used. Once we receive the completed questionnaire, we will look at the protocol and see if there are any suggestions we can make that may improve the results.

    Vielen Dank für Ihren Anruf.

    Wie am Telefon besprochen, werden die Mandeln viel als positive Kontrolle für humanes Gewebe genommen. In der Maus wird viel ein Embryo, Hirn oder Retina auch benutzt. Wie aber auch am Telefon angesprochen, sollten die Lymphknoten ja auch aktivierte Caspase 3 positive Zellen haben.

    I'm having trouble with this antibody as it is giving me a lot in terms of what I believe to be non-specific staining, cells fluorescing (colocalize with DAPI) in 5um sections of parrifin embedded, formalin fixed, brain sections of naive mice, but they are not neurons (do not colocalize with non-specific neuronal marker). Have there been any complaints of impurities in this particular lot that may be causing this reaction? We have attemtpted caspase 3 before, but in mouse not rat and have been successful. I've tried several different protocols, any ideas of what is going on? Again, there is reactivity, but in naive mice as well. blocking condition: 10% goat/1% BSA 20-60 minutes (tried, 20, 40, 60) permeabilization with TX-100 0.3% 60 minutes detection: tried LSAB and fluorescence (alexa 488) antigen retrieval (heat based sodium citrate) 2ndary (alexa 488, 1:250-1:1000 tried) 1ary (1:20-1:100 tried) wash: PBS 5x3 min between antibodies parafin embeded sections, fixed with PFA: brain sample

    Thank you for sending your protocol details. We have not received other complaints about this lot, but I do see one complaint from February about a different lot but with a different issue: no staining at all.

    If I understand you correctly, you expect to see activated caspase 3 in neurons, in both groups of mice, naive and treated. Do you have any other evidence that the treatment is inducing apoptosis in neurons? Can you please send an image of the stain?

    Are you confident your secondary antibody and reagents are not the source of the stain? A no-primary-antibody control will confirm, if this is the case.

    Thank you for contacting us yesterday and your interest in our products.

    I have had a look at the reference we discussed yesterday, mentioned in the specificity section of the anti-active Caspase 3 antibody (ab2302):

    Bhoopathi P et al. Blockade of tumor growth due to matrix metalloproteinase-9 inhibition is mediated by sequential activation of 1-integrin, ERK, and NF-kappaB. J Biol Chem 283:1545-52 (2008).PubMed: 17991734

    This reference is specifically mentioned as it has used the antibody and detected both the pro and active forms of Caspase 3. This is in contrast to the conditions which we have used in our Western blotting experiments and several other references where only the p17 active Caspase 3 fragment was detected.

    The mention of this reference is simply to illustrate that under certain experimental conditions the pro form may be detected, but we have found the antibody to preferentially recognise the pro-form.
