    Caspases are synthesized as inactive pro-enzymes that are processed to active form in cells undergoing apoptosis. Caspase 7 has been identified as a major contributor to the execution of apoptosis. Caspase 7 is cleaved by many enzymes, including caspase 3, 6, 8, 9 and granzyme B. Active caspase 7 consists of a heterotetramer of two large and two small subunits. As an effector caspase, caspase 7 cleaves many of the same substrates as caspase 3, including poly (ADP-ribose) polymerase (PARP). The active caspase 7 antibody provides a new tool for identifying apoptotic cell populations in both tissue sections and cultured cells.

  • Relevance
    Caspases are cysteine proteases, expressed as inactive precursors, that mediate apoptosis by proteolysis of specific substrates. Caspases have the ability to cleave after aspartic acid residues. There are two classes of caspases involved in apoptosis; initiators (activation by receptor cluster) and effectors (activation by mitochondrial permeability transition). Proapoptotic signals autocatalytically activate initiator caspases, such as Caspase 8 and Caspase 9. Activated initiator caspases then process effector caspases, such as Caspase 3 and Caspase 7, which in turn cause cell collapse.

I'm sorry to hear you are having a problem with ab2323 I would like to suggest the following modifications to your protocol: block for 1 hour at room temperature in 5% milk, incubate the primary at 1 ug/mL in blocking buffer overnight at 4 degrees, and incubate in secondary in blocking buffer for 1 hour at room temp. These are the conditions for this antibody. An image of the Western blot for this antibody has been added to the datasheet on the website. The antibody at 1 ug/mL detected active caspase-7 in Jurkat cells induced with 2 mM of Camptothecin for 6 hours. I can tell you that we did not soucre this antibody from cell signalling but our suppliers names are commercially sensitive information. The antibody should be used at 10-20ug/ml in frozen sections and antigen retrieval is not required although it may enhance staining.
