General notes
ab256165 is the carrier-free version of ab222408.
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. The carrier-free buffer and high concentration allow for increased conjugation efficiency.
This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.
Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with <1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.
This product is a recombinant monoclonal antibody, which offers several advantages including:
- - High batch-to-batch consistency and reproducibility
- - Improved sensitivity and specificity
- - Long-term security of supply
- - Animal-free production
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

Target
-
Function
Plays a role in the TP53-regulated cellular response to DNA damage probably by cooperating with TP53 (PubMed:16897187, PubMed:18223688, PubMed:21522185). The N-terminal moiety promotes pyroptosis and exhibits bactericidal activity (PubMed:27281216). The physiological relevance of these observations is unknown. -
Tissue specificity
Expressed in cochlea. Low level of expression in heart, brain, placenta, lung, liver, skeletal muscle, kidney and pancreas, with highest expression in placenta. -
Involvement in disease
Deafness, autosomal dominant, 5
Is a tumor suppressor gene with an important role in colorectal cancer (CRC). -
Sequence similarities
Belongs to the gasdermin family. -
Domain
Intramolecular interactions between N- and C-terminal domains may be important for autoinhibition in the absence of activation signal. The intrinsic pyroptosis-inducing activity is carried by the N-terminal domain. -
Cellular localization
Cytoplasm, cytosol. Cell membrane. - Information by UniProt

All lanes : Anti-cleaved N-terminal DFNA5 / GSDME antibody [EPR20867-248] (ab222408) at 1/1000 dilution
Lane 1 : Wild-type SH-SY5Y (human neuroblastoma cell line from bone marrow) (treated with treated with etoposide (60uM, 14h), whole cell lysate
Lane 2 : Untreated wild-type SH-SY5Y whole cell lysate
Lane 3 : DFNA5/GSDME knockout SH-SY5Y treated with etoposide (60 uM, 14h), whole cell lysate
Lysates/proteins at 20 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/20000 dilution
Predicted band size: 55 kDa
Exposure time: 27 secondsBlocking and dilution buffer: 5% NFDM/TBST.
The lysates were kindly provided by our collaborator Dr Feng Shao's lab, NIBS.
The molecular weight observed is consistent with the literature (PMID: 28459430, 30804337).
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab222408).
