Target

  • Function

    C-type lectin that functions as cell-surface receptor for a wide variety of ligands such as damaged cells, fungi and mycobacteria. Plays a role in the recognition of pathogenic fungi, such as Candida albicans. The detection of mycobacteria is via trehalose 6,6'-dimycolate (TDM), a cell wall glycolipid. Specifically recognizes alpha-mannose residues on pathogenic fungi of the genus Malassezia. Recognizes also SAP130, a nuclear protein, that is released by dead or dying cells. Transduces signals through an ITAM-containing adapter protein, Fc receptor gamma chain /FCER1G. Induces secretion of inflammatory cytokines through a pathway that depends on SYK, CARD9 and NF-kappa-B.

  • Paraffin-embedded human testis tissue stained for CLEC4E/MINCLE using ab236605 at 1/600 dilution in immunohistochemical analysis.

    After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

  • Paraffin-embedded human kidney tissue stained for CLEC4E/MINCLE using ab236605 at 1/600 dilution in immunohistochemical analysis.

  • U-251 MG (human brain glioma cell line) cells stained for CLEC4E/MINCLE (green) using ab236605 at 1/250 dilution in ICC/IF. Secondary antibody is Alexa Fluor® 488-conjugated Goat Anti-Rabbit IgG (H+L). Counter-stained with DAPI (blue).

    The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C.
