Function
Functions as an endocytic receptor on a small subset of myeloid cells specialized for the uptake and processing of material from dead cells. Recognizes filamentous form of actin in association with particular actin-binding domains of cytoskeletal proteins, including spectrin, exposed when cell membranes are damaged, and mediate the cross-presentation of dead-cell associated antigens in a Syk-dependent manner. -

This data was developed using ab300433, the same antibody clone in a different buffer formulation.
Immunohistochemical analysis of paraffin-embedded mouse spleen tissue labelling CLEC9A with ab300433 at 1/100 followed by a ready to use LeicaDS9800 (BOND™ Polymer Refine Detection). Membraneous staining on mouse spleen is observed. The section was incubated with ab300433 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.
Secondary antibody only control: PBS was used instead of primary antibody followed by ready to use secondary antibody LeicaDS9800 (BOND™ Polymer Refine Detection).

Intracellular flow cytometric analysis of 2% paraformaldehyde fixed and 0.1% Tween-20 permeabilized mouse splenocytes labelling CLEC9A with ab300433 at 1/50 dilution (0.1µg) (Right) compared with a Rabbit monoclonal IgG (ab172730) (Left) isotype control. Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/2000 dilution was used as the secondary antibody.
Cells were stained with anti-CD11c conjugated to PE-Cy5.5 and anti-CD8a conjugated to Alexa Fluor® 647.
Gated on CD11c+ cells.
