Thank you for your patience. None of our CLK antibodies have been tested against purified protein of other CLK family members. Cross-reactivity of, for instance, a CLK1 antibody with CLK2, 3, or 4 has not been confirmed. The best we can do is predict the potential for cross-reactivity by aligning the immunogens of each antibody, when they are available, with the sequences of the other proteins. The N terminus appears to have a lot of diversity among these four, so antibodies raised against the N terminus are likely to have specificity for their targets. For example, the CLK2 antibody ab86147 is raised against the CLK2 N terminus which has little or no homology with the N termini of CLK1, 3, or 4. This is also the case for the CLK3 antibody ab37919, raised against the N terminus of CLK3. We do not have the immunogen sequence information for the CLK1 and CLK4 antibodies in our catalogue, but I can obtain alignment scores for these if you are still interested. For proteins, we only have a CLK1 protein, ab114706, full-length recombinant human with a 26 kDa GST tag. There is also a CLK2 protein fragment, but it does not include the N terminus. I performed a BLAST to find out the percentage of homology between the four proteins, and the results are: -CLK1-CLK2: 53.5% -CLK1-CLK3: 14% -CLK1-CLK4: 77% -CLK2-CLK3: 17% -CLK2-CLK4: 55% -CLK3-CLK4: 14% We usually predict cross-reactivity when the homology between the sequences is over 85%. So, in this case, I would dare to affirm there might be some slight reactivity between CLK1 and CLK4, but the rest of proteins are very unlikely to cross react.
