Fresh frozen tissues were cut at 10 microns, then mounted onto Superfrost Plus slides.
After drying overnight in a working Fume hood, sections were fixed in 4% Formalin in PBS for 10 mins, before immunostaining.
It seems to identify small fibres very well.
However, I would have liked to have been able to use a lower dilution factor.
NB: by using a biotinylated secondary and then incubating in avidin-Fluorochrome, you will be able to dilute the primary Ab to at least 1/200.
