Properties

Applications

Our Abpromise guarantee covers the use of ab62590 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1.25 µg/ml. Detects a band of approximately 45 kDa (predicted molecular weight: 40 kDa). Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
IHC-P Use a concentration of 4 - 8 µg/ml.

Target

  • Function
Binds and transactivates the retinoic acid response elements that control expression of the retinoic acid receptor beta 2 and alcohol dehydrogenase 3 genes. Transactivates both the phenobarbital responsive element module of the human CYP2B6 gene and the CYP3A4 xenobiotic response element.
  • Tissue specificity
    Predominantly expressed in liver.
  • Sequence similarities
Belongs to the nuclear hormone receptor family. NR1 subfamily.
Contains 1 nuclear receptor DNA-binding domain.
  • Domain
Composed by a short N-terminal domain followed by the DNA binding, hinge, and ligand binding/dimerization domains.
  • Post-translational
    modifications
Phosphorylated at Thr-38 by PKC, dephosphorylation of Thr-38 is required for nuclear translocation and activation.
  • Cellular localization
Nucleus. Cytoplasm. Cytoplasm > cytoskeleton. Recruited to the cytoplasm by DNAJC7.

Answer

Thank you for your inquiry.
After reviewing the IHC image, I can provide the following answers:
1. Other cells besides the 2 marked with arrows are positively stained; the arrows show nice examples of positively stained hepatocytes.
2. The best IHC image we have is on the website and I am sorry to confirm that we cannot provide one with a better resolution.
3. It is difficult to see nuclear staining because of the counterstain that marks the nuclei in blue. We can see cytoplasmic and nuclear staining. which is consistent with Uniprot localization data (http://www.uniprot.org/uniprot/Q14994).
4. When blasting the exact immunogen sequence used to produce this antibody, I found homology with isoforms 2, 3, and 7. Isoforms 1, 2 and 3 have similar molecular weight, and isoform 7 is only a little shorter. No detectable band for isoform 7 may be due to limit of detection issues.
I hope this information is helpful and wish you and your customer a good week too.

Question
Answer

Thank you for taking time to complete our questionnaire. I am sorry to hear that this antibody is not providing satisfactory results.

The details provided will enable us to investigate this case and will provide us with vital information for monitoring product quality.

Having reviewed this case,I would also appreciate if you can confirm some further details to help optimise the results from ab62590 Anti-Constitutive androstane receptor antibody:

1. What kind of lysis buffer did you use?

2.Do you use a reducing loading buffer?

3. Is you gel run under reducing conditions?

4. Did you run a non-primary control?

5. Is it possible to send us an image?

In the event that a product is not functioning in the species and applications cited on the product datasheet (and the problem has been reported within 6 months of purchase), we would be pleased to provide a free of charge replacement, credit note, or refund.

I hope this information is helpful, and I thank you for your cooperation.
