Vimentin is a type III intermediate filament protein with a molecular weight of ~54kDa. Vimentin anchors and supports organelles within the cytosol of mesenchymal cells and is upregulated during epithelial to mesenchymal transition (EMT), a process that occurs during neural development, wound healing, and cancer metastasis. Vimentin staining can be used as a marker for mesenchymal-derived cells, or cells undergoing EMT. Due to its diverse role within the cell, vimentin can be studied in many contexts ranging from cancer to cytoskeletal dynamics.
During metastasis, cancer cells can often obtain characteristics such as EMT, which vimentin contributes to by changing cell shape and motility. Therefore, immunotherapy targeting phosphorylated vimentin is showing potential in the treatment of metastatic tumors while sparing non-tumor cells1. Studies related to proteomics have shown that vimentin is a metastasis-associated factor in multiple malignancies, such as prostate and breast cancer. This suggests it could play an important role in tumor progression and serve as a potential biomarker for metastasis2.

|||Immunohistochemical staining of paraffin embedded human cervical carcinoma with purified ab92547 at a working dilution of 1/250. The secondary antibody used is HRP goat anti-rabbit IgG H&L (ab97051) at 1/500. The sample is counter-stained with hematoxylin. Antigen retrieval was perfomed using Tris-EDTA buffer, pH 9.0. PBS was used instead of the primary antibody as the negative control, and is shown in the inset.|

ab8978 staining Vimentin in wild-type HAP1 cells (top panel) and VIM knockout HAP1 cells (bottom panel). The cells were fixed with 100% methanol (5min), permeabilized with 0.1% Triton X-100 for 5 min and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated with ab8978 at 1μg/ml and ab202272 (Rabbit monoclonal [EP1332Y] to alpha Tubulin (Alexa Fluor® 594)) at 1/250 dilution overnight at +4°C, followed by a further incubation at room temperature for 1h with ab150117 (Goat secondary antibody to Mouse IgG (Alexa Fluor® 488)) at 2 μg/ml (shown in green). Nuclear DNA was labelled in blue with DAPI.
