  • Product overview

Abcam’s Cortisol in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Cortisol in saliva.

A 96-well plate has been precoated with anti-Cortisol IgG. Samples and the Cortisol-HRP conjugate are added to the wells, where Cortisol in the sample competes with the added Cortisol-HRP for antibody binding. After incubation, the wells are washed to remove unbound material and TMB substrate is then added which is catalyzed by HRP to produce blue coloration. The reaction is terminated by addition of Stop Solution which stops the color development and produces a color change from blue to yellow. The intensity of signal is inversely proportional to the amount of Cortisol in the sample and the intensity is measured at 450 nm.

Happy with the product

Good Excellent 5/5 (Ease of Use)
We bought this kit mainly because it does not cross react with dexamethasone as stated in the protocol. We used guinea pig saliva and followed the protocol exactly. Overall, kit was easy to use with straight forward instructions. Our curve was good, all samples fell within the range as expected. Only reason not rating this a 5* is that although the kit should not detect dex our own internal controls contradicted this, but only slightly and the dex was a high concentration. Will use this kit again.

For ab154996 we recommend to collect saliva samples with a centrifuge glass tube and a plastic straw or with the “Salivette” (Sarstedt, Ref. 511534500).

The following should be noted:
a) If saliva collection is carried out in the morning ensure that this is carried out prior to brushing teeth
b) During the day allow 1 hour after any food or drink before collecting saliva samples
c) It is very important that a good clear sample is received – i.e. no contamination with food, lipstick, blood
(bleeding gums) or other extraneous materials.

Processing Instructions
1. Let the saliva flow down through the straw into the centrifuge glass tube
2. Centrifuge at 3000 rpm for 15 min.
3. Store at -20°C for at least 1 hour
4. Defrost samples
5. Centrifuge again for 15 min. at 3000 rpm
6. The saliva sample is now ready to be tested.
7. Store the sample at 4 °C for one week or at -20°C for longer term storage.

Saliva Processing Instructions with Salivette Sarstedt
1. Remove the swab from the suspended insert of the Salivette.
2. Gently chewing the swab for 1 minute produces a sufficient quantity of Saliva.
3. Replace the swab into the Salivette and firmly close the tube using the stopper.
4. Centrifuge the Salivette for 2 minutes at 1000g (rcf) for saliva generation.
5. Remove the insert complete with the swab from the centrifuge vessel and discard. The clear Saliva is now
ready for analysis (at least 1 ml of saliva should be recovered with this method).

For storage of samples we would recommend to freeze them at -20C. We have not tested how long samples can be stored frozen without loosing quality but believe a couple month should not be a problem.
