    Cre-recombinase is a 38 kDa DNA recombinase derived from the P1 bacteriophage. It is highly specific for a 34 bp DNA sequence (loxP) found in P1 DNA. It catalyzes site-specific recombination between two 34-base-pair LOXP sites. Its role is to maintain the phage genome as a monomeric unit-copy plasmid in the lysogenic state. It is homotetramer when bound to DNA and belongs to the "phage" integrase family.

Question

DESCRIPTION OF THE PROBLEM No signal at all. With 2 different western blot experiments, we obtained no band corresponding to the size of the Cre recombinase (but a lot of non-specific bands). We used 2 different types of samples: first we loaded proteins extracted from different organs of a Nestin-Cre mouse in addition to a positive control (HEK cells transfected with a CMV-Cre plasmid). With the second experiment, we have loaded protein lysate from cardiac fibroblasts infected either with an adenovirus CMV-Cre or a CMV-LacZ. The transduction worked as I could detect a specific band for Beta-galactosidase on my blot but no band at all for the Cre. SAMPLE - protein lysate from different organs of Nestin_Cre mouse - protein lysate from fibroblasts infected with a CMV-Cre adenovirus. PRIMARY ANTIBODY Anti Cre recombinase antibody from Abcam.dilution 1/200 and 1/100. Incubation overnight 4 degrees. DETECTION METHOD ECLPlus POSITIVE AND NEGATIVE CONTROLS USED Nestin-Cre mouse protein lysate and HEK cells transfected with CMV-Cre plasmid ANTIBODY STORAGE CONDITIONS -20 degrees and aliquoted at delivery. SAMPLE PREPARATION Sample were prepared using the M-PER buffer from Thermo. Protease and phosphatase inhibitors were added and samples diluted in Laemmli buffer were boiled for 5 minutes. AMOUNT OF PROTEIN LOADED - 40 microg ELECTROPHORESIS/GEL CONDITIONS Reducing gel, 8% gel TRANSFER AND BLOCKING CONDITIONS Blocking in either 5% milk or 5% BSA (for the fibroblast samples). SECONDARY ANTIBODY Goat anti-rabbit diluted at 1/3000 HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 4 HAVE YOU RUN A "NO PRIMARY" CONTROL? No DO YOU OBTAIN THE SAME RESULTS EVERY TIME? Yes WHAT STEPS HAVE YOU ALTERED? We tried different blocking solution (milk or BSA) and tried 1/200 dilution (as recommended by Abcam. Actually a very high concentration)and 1/100 dilution. ADDITIONAL NOTES I actually understand that other labs faced the same problem as us with this antibody for Western blot. It sounds to me that this Ab is not suitable for Western blot. Thanks for your help.

Thank you for bringing this to our attention and for completing the troubleshooting form. Your sample preparation and protocol are correct. I agree that the antibody appears to be the problem. We do not have a different lot, so I suggest replacing this with a different Cre recombinase antibody, if you are interested. If not, I can issue a Credit or refund. Alternatively, if you are interested in an antibody against a different protein, I can send that as a replacement, instead of a Cre antibody.

The two other Cre recombinase antibodies we have are ab41104 and ab101071.
