General notes
Crystal Violet Solution ab246820 is the standard dye for staining gram positive bacteria.
Gram positive bacteria: Blue.
Control Tissue: Any well fixed tissue section. Air dried smear.
Procedure:
1. Deparaffinize sections if necessary and hydrate to distilled water.
2. Apply adequate Crystal Violet Solution to completely cover tissue section and incubate for 1-3 minute(s).
3. Rinse slide in distilled water to remove excess stain.
4. Continue with desired gram staining protocol.
Note: Gram positive bacteria that are dying, dead or being treated with antibiotics may stain variably (red).

Relevance
Gram-positive bacteria are characterized by their blue-violet colour reaction in the Gram-staining procedure. The colour reaction is caused by crystal-violet, the primary Gram-stain dye, complexing with the iodine mordant. When the decolorizer is applied, a slow dehydration of the crystal-violet/iodine complex is observed due to the closing of pores running through the cell wall. -
