  • General notes

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Target

  • Function

Essential component of the COP9 signalosome complex (CSN), a complex involved in various cellular and developmental processes. The CSN complex is an essential regulator of the ubiquitin (Ubl) conjugation pathway by mediating the deneddylation of the cullin subunits of SCF-type E3 ligase complexes, leading to decrease the Ubl ligase activity of SCF-type complexes such as SCF, CSA or DDB2. The complex is also involved in phosphorylation of p53/TP53, c-jun/JUN, IkappaBalpha/NFKBIA, ITPK1 and IRF8/ICSBP, possibly via its association with CK2 and PKD kinases. CSN-dependent phosphorylation of TP53 and JUN promotes and protects degradation by the Ubl system, respectively. Suppresses G-protein-and mitogen-activated protein kinase-mediated signal transduction.
  • Tissue specificity

    Widely expressed.
  • Sequence similarities

    Belongs to the CSN1 family.
    Contains 1 PCI domain.
  • Domain

The PCI domain is necessary and sufficient for the interactions with other CSN subunits of the complex. Mediates the interaction with CAPN8.
The N-terminal part (1-216), which is not required for deneddylating activity and CSN complex formation, is nevertheless essential for other aspects of CSN complex function, such as repression of c-fos/FOS expression.
  • Post-translational
    modifications

Phosphorylated upon DNA damage, probably by ATM or ATR.
  • Cellular localization

    Cytoplasm. Nucleus.

Question

I was looking up the CSN1 antibody you described has been synthesized according to the exon sequence between exon 13 and 14 of the respective human gene. Now we would like to use this antibody against an ortholog in Aspergillus nidulans a filamentous fungus. Since my experience with antibodies is limited I would greatly appreciate if you could evaluate our chances on how good this antibody would work against our protein. To give you some more information, this is the genbank accession number where you can find the information about coding sequences and predicted amino acid sequences: AAT73769 Also it would be of interest as to where the antibody would most likely bind, since I was unable to follow the locus link ID you provided, an indication showing us the involved amino acid region would be very helpful. Thank you very much for your support. With kind regard Oliver Draht

Answer

Thank you for your enquiry. This product will not work for E. nidulans. The epitope recognized by ab10413 maps to a region between residue 400 and the C-terminus (residue 471) of human G protein Pathway Suppressor 1 using the numbering given in Swiss-Prot entry Q13098 (GeneID 2873). I hope this information helps. Please do not hesitate to contact us if you need anything further.
