Our Abpromise guarantee covers the use of ab76470 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB 1/2000 - 1/10000. Detects a band of approximately 88, 102 kDa (predicted molecular weight: 102 kDa).
IHC-P 1/100 - 1/250. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.

Use of HRP-conjugated or polymerized HRP secondary antibodies, stronger signals have been found using the polymerized HRP secondary.

Flow Cyt 1/60.

ab172730 - Rabbit monoclonal IgG, is suitable for use as an isotype control with this antibody.

  • Application notes
Is unsuitable for ICC/IF or IP.
  • Target

    • Relevance

Core component of multiple cullin-RING-based E3 ubiquitin-protein ligase complexes which mediate the ubiquitination and subsequent proteasomal degradation of target proteins. As a scaffold protein may contribute to catalysis through positioning of the substrate and the ubiquitin-conjugating enzyme. The E3 ubiquitin-protein ligase activity of the complex is dependent on the neddylation of the cullin subunit and is inhibited by the association of the deneddylated cullin subunit with TIP120A/CAND1. The functional specificity of the E3 ubiquitin-protein ligase complex depends on the variable substrate recognition component.

    Customer reviews and Q&As

    1-2 of 2 Abreviews or Q&A

Thank you for your enquiry regading ab76470.
I can confirm that the sequence we used to raise Anti-Cullin 4A/B antibody maps to a region that is conserved between CUL4A and B.
In addition, we observe 2 bands (at 88 and at 102 kDa) in samples that express both isoforms.
    - Have you tested a sample that is known to express CULA, such as A375 or HeLa?
I have attached an additional image showing the CUL4A expression in mouse and rat samples to share with you.
If you need any further assistance in the future, please do not hesitate to contact me.
