Our Abpromise guarantee covers the use of ab65968 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 µg/ml. Detects a band of approximately 41 kDa (predicted molecular weight: 41 kDa).
IHC-P Use a concentration of 5 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

  • Function

Receptor for interleukin-8 which is a powerful neutrophil chemotactic factor. Binding of IL-8 to the receptor causes activation of neutrophils. This response is mediated via a G-protein that activates a phosphatidylinositol-calcium second messenger system. Binds to IL-8 with high affinity. Also binds with high affinity to CXCL3, GRO/MGSA and NAP-2.
  • Sequence similarities

Belongs to the G-protein coupled receptor 1 family.
  • Post-translational

Phosphorylated upon ligand binding; which is required for desensitization.
  • Cellular localization

    Cell membrane.

  • IHC image of ab65968 staining in human breast carcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab65968, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Thank you for submitting an Abreview for ab25968. We greatly appreciate your feedback and strive to publish both negative and positive reviews, rewarding each with Abpoints. Before we publish negative Abreviews our Scientific Support team reviews each one. I am wondering whether the faint band is in fact the CXCR2 band or the stronger band a bit higher. Often bands will appear a few kD off from the projected size based on modifications, or even gel and buffer type. You may wish to try another cell type that is CXCR2 decifient for comparison. We do guarantee all our products for the tested species and application within 6 months of purchase. If you are unhappy with your results within this time frame please send us your order number and we can offer a free of charge replacement, credit or refund. Thank you very much for your time and we look forward to your reply.
