Target

FunctionReceptor for CXCL9, CXCL10 and CXCL11 and mediates the proliferation of human mesangial cells (HMC).
Isoform 2 is a receptor for CXCL4 and also mediates the inhibitory activities of CXCL9, CXCL10 and CXCL11 on the growth of human microvascular endothelial cells (HMVEC). May play a role in angiogenesis.
Isoform 3 mediates the activity of CXCL11. -
Tissue specificityIsoform 1 and isoform 2 are mainly expressed in heart, kidney, liver and skeletal muscle. Isoform 1 is also expressed in placenta.

Immunocytochemistry/ Immunofluorescence - Anti-CXCR3 antibody [49801] (ab64714)This image is courtesy of an Abreview submitted by Bing Wangab64714 staining CXCR3 in primary cultured HUVEC by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with acetone and blocked with 5% serum for 30 minutes at 24°C. Samples were incubated with primary antibody (1/400 in PBS + 2.5% serum) for 16 hours at 4°C. An Alexa Fluor®488-conjugated Goat anti-mouse IgG polyclonal (1/500) was used as the secondary antibody.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-CXCR3 antibody [49801] (ab64714)This image is courtesy of an Abreview submitted by Frank Whiteab64714 staining CXCR3 in pig uterus tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded tissue sections). The sections were fixed in paraformaldehyde and subjected to enzymatic antigen retrieval prior to blocking with 10% serum for 20 minutes at 24°C. The primary antibody was diluted 1/200 and incubated with the sample for 30 minutes at 24°C. A biotin-conjugated ABC kit was used to detect the antibody.

The chemoattractive activity of recombinant human CXCL11 (7 ng/ml) towards B cells transfected with human CXCR3 is neutralized in a dose-dependent manner by ab64714 (green). The IC50 for neutralization is typically 0.3 - 1.5 μg/ml.

Flow Cytometry of HEK293 cells transfected with human CXCR3 (upper histogram) or with irrelevant transfectants and eGFP (lower histogram) were stained with ab64714(10μl undiluted antibody per 1 x106 cells) followed by APC conjugated anti-mouse IgG secondary antibody.

The sections were fixed in formaldehyde and subjected to heat-mediated antigen retrieval in citrate buffer prior to blocking with a proprietary non-protein reagent for 5 minutes at 25°C. The primary antibody was diluted 1/2000 and incubated with the sample for 30 minutes. A biotin-conjugated goat anti-mouse IgG was used as the secondary antibody.
