Target

Images

  • ab3563 at 1/300 dilution detecting 0.5ug recombinant human cyclophilin A by Western Blot (ECL).

  • ICC/IF image of ab3563 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab3563, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

  • All lanes : Anti-Cyclophilin A antibody (ab3563) at 1/500 dilution

Lane 1 : HeLa cell lysate
    Lane 2 : K562 cell lysate
    Lane 3 : HepG2 cell lysate
    Lane 4 : NIH-3T3

Lysates/proteins at 25 µg per lane.

    Observed band size: 18 kDa
why is the actual band size different from the predicted?

Answer

Thank you for your enquiry. This product ab3563 is supplied as 400 µl of total IgG in PBS containing 0.05% sodium azide. Unfortunately, the concentration of this product has not been determined at this time, as the antibody has only been protein (and not epitope affinity) purified. I am sorry I can not be of more assistance in this occasion but should you require any further help please do not hesitate to contact me.

Thank you for your enquiry. This antibody is listed on the data sheet as being an IgG fraction. Because this antibody is not affinity purified, the concentration is undetermined. I understand that you need the concentration for your assays, but unfortunately that information is unattainable. I apologize for any inconveniance this has caused you.

Question
Answer

Thank you for your enquiry. Regarding polyclonal antibody to Cyclophilin A ab3563, our source has not measured the IgG concentration. The activity of some of the antibodies we sell is based on ELISA data. However, the concentration is expected to be between 1-2 mg/ml. I apologize for any inconvenience this may cause. Please let me know if I may provide further assistance.

Answer

Thank you for your enquiry. Cyclophilin A is described as a housekeeoping gene in the literature. However, this cyclophilin A antibody does not detect endogenous levels of CyPA, so it would not be suitable for this purpose. I hope this information helps, please do not hesitate to contact us if you need any more advice or information.

Answer

I apologize, but it looks like the IgG concentration is unknown for ab3563 (Cyclophilin A). Occasionally, ELISA tests are used to determine activity instead of measuring the protein concentration. More than likely, the range falls between 0.5-2 mg/ml. Please contact us again if you have any additional questions.
