Target

  • Function

    Cytochromes P450 are a group of heme-thiolate monooxygenases. In liver microsomes, this enzyme is involved in an NADPH-dependent electron transport pathway. It oxidizes a variety of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. In the epoxidation of arachidonic acid it generates only 14,15- and 11,12-cis-epoxyeicosatrienoic acids. It is the principal enzyme responsible for the metabolism the anti-cancer drug paclitaxel (taxol).

    Question

    In reference to our telephonic conversation about Anti-CYP2C8 antibody (ab88904) I would like to inquire about the antibody reactivity with different truncated products which I plan to make with original (Wild Type) CYP2C8 which is protein sequence of 490 amino acid in length. These truncated products are the variants I plan to detect cytochrome P450 2C8 isoform b [Homo sapiens] 420 aa protein and also cytochrome P450 2C8 isoform c [Homo sapiens] 388 aa protein using this antibody.
    I was wanting to know if this antibody would work good for detection of this variants, since these variants are N- terminal truncated products, so if the antibody is generated to the N-terminal end of the protein it think it can give me problems. For better understanding of the sequence these variants share, I am attaching the sequence similarity match as attachment, Please have take a look.
    Isoform 1 is WT;
    Isoform 2 is cytochrome P450 2C8 isoform b;
    Isoform 3 is cytochrome P450 2C8 isoform c
