Function
Catalyzes the last step in the transsulfuration pathway from methionine to cysteine. Has broad substrate specificity. Converts cystathionine to cysteine, ammonia and 2-oxobutanoate. Converts two cysteine molecules to lanthionine and hydrogen sulfide. Can also accept homocysteine as substrate. Specificity depends on the levels of the endogenous substrates. Generates the endogenous signaling molecule hydrogen sulfide (H2S), and so contributes to the regulation of blood pressure. -

Involvement in disease
Defects in CTH are the cause of cystathioninuria (CSTNU) [MIM:219500]. It is an autosomal recessive phenotype characterized by abnormal accumulation of plasma cystathionine, leading to increased urinary excretion. -

Lane 1: Wild-type HAP1 whole cell lysate (20 µg)
Lane 2: Cystathionase/CTH knockout HAP1 whole cell lysate (20 µg)

Lanes 1 - 4: Merged signal (red and green). Green - ab189916 observed at 45 kDa. Red - loading control, ab130007, observed at 130 kDa.
ab189916 was shown to recognize Cystathionase/CTH in wild-type cells as signal was lost at the expected MW in Cystathionase/CTH knockout cells. Additional cross-reactive bands were observed in the wild-type and knockout cells. Wild-type and Cystathionase/CTH knockout samples were subjected to SDS-PAGE. ab189916 and ab130007 (Mouse anti-Vinculin loading control) were incubated overnight at 4°C at 1/10000 dilution and 1/20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

Immunoprecipitation of Cystathionase/CTH from HeLa cell lysate using ab189916 at 1/50 or 1X PBS (negative control) (lane 1). Immunostained with ab189916 at 1/1500 and HRP-conjugated anti-rabbit IgG preferentially detecting the non-reduced form of rabbit IgG.
