• Product name

    Anti-Cytokeratin 1 + 5 + 10+ 14 antibody [34(beta)E12]
  • Description

    Mouse monoclonal [34(beta)E12] to Cytokeratin 1 + 5 + 10+ 14
  • Host species

  • Tested applications

    Suitable for: IHC-P, WBmore details
  • Species reactivity

    Reacts with: Human
  • Immunogen

Tissue, cells or virus corresponding to Human Cytokeratin 1 + 5 + 10+ 14. Keratin solubilized from Human stratum corneum.

  • Positive control

    • ab195381 reacts with all squamous and ductal epithelium as well as carcinomas such as squamous cell carcinoma and adenocarcinoma of ductal origin (e.g., breast, pancreas) but does not react with sarcoma, melanoma or lymphoma. ab195381 reacts with benign small-acinar lesions of the prostate and has been shown to be useful as an adjunct in the identification of prostate carcinoma.
  • General notes

ab195381 is specific for "high molecular weight" cytokeratins 1, 5, 10 and 14, corresponding to molecular weights 68, 58, 56.5 and 50 kDa, respectively, which are characteristically found in "complex" epithelium.

Application Abreviews Notes
IHC-P 1/20.

For optimal results, fixation in alcohol-based fixatives, such as methacarn, is recommended. Pretreatment with proteolytic enzymes such as trypsin may be required when formalin-fixed sections are to be stained. For optimal staining of prostate tissue higher concentrations of protease and longer incubation times are recommended, e.g., 0.5% trypsin at 37°C for 2 to 4 hours.

WB 1/20 - 1/50.

  • Function

May regulate the activity of kinases such as PKC and SRC via binding to integrin beta-1 (ITB1) and the receptor of activated protein kinase C (RACK1/GNB2L1). In complex with C1QBP is a high affinity receptor for kininogen-1/HMWK.
  • Tissue specificity

The source of this protein is neonatal foreskin. The 67-kDa type II keratins are expressed in terminally differentiating epidermis.
  • Involvement in disease

    Epidermolytic hyperkeratosis
    Ichthyosis hystrix, Curth-Macklin type
    Keratoderma, palmoplantar, non-epidermolytic
    Ichthyosis annular epidermolytic
    Keratoderma, palmoplantar, striate 3
  • Sequence similarities

Belongs to the intermediate filament family.
  • Post-translational

Undergoes deimination of some arginine residues (citrullination).
  • Cellular localization

Cell membrane. Located on plasma membrane of neuroblastoma NMB7 cells.
