FunctionPlays a significant role in maintaining keratin filament organization in intestinal epithelia. When phosphorylated, plays a role in the secretion of mucin in the small intestine.
Tissue specificityExpressed predominantly in the intestinal epithelium. Expressed in luminal cells of colonic mucosa. Also expressed in the Merkel cells of keratinized oral mucosa; specifically at the tips of some rete ridges of the gingival mucosa, in the basal layer of the palatal mucosa and in the taste buds of lingual mucosa.
Sequence similaritiesBelongs to the intermediate filament family.
Developmental stageFirst detected at embryonic week 8 in individual 'converted' simple epithelial cells of the developing intestinal mucosa. In later fetal stages, synthesis extends over most goblet cells and a variable number of villus enterocytes. In the developing gastric and intestinal mucosa, expressed in all enterocytes and goblet cells as well as certain 'low-differentiated' columnar cells, whereas the neuroendocrine and Paneth cells are negative.
modificationsHyperphosphorylation at Ser-13 occurs during the early stages of apoptosis but becomes less prominent during the later stages. Phosphorylation at Ser-13 also increases in response to stress brought on by cell injury.
Proteolytically cleaved by caspases during apoptosis. Cleavage occurs at Asp-228.

Formalin-fixed, paraffin-embedded human colon tissue stained for Cytokeratin 20 with ab230524 at a 1:100 dilution in immunohistochemical analysis.
All lanes : Anti-Cytokeratin 20 antibody - C-terminal (ab230524) at 1/500 dilution
Lane 1 : A549 (Human lung carcinoma cell line) whole cell lysate
Lane 2 : NS-1 whole cell lysate
Lane 3 : Mouse kidney whole cell lysate
Lane 4 : Rat kidney whole cell lysate
Developed using the ECL technique.
Predicted band size: 48 kDa
A549 (Human lung carcinoma cell line) cells stained for Cytokeratin 20 using ab230524 at a dilution of 1/100 in ICC/IF.
Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with ab230524 in 3% BSA-PBS and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight® 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
