Answer

Answer

Thank you for confirming that information.

Kate is currently away from the office but I will try to assist you as best I can.

As I am sure you are aware, the antibody purchased, Anti-Cytokeratin 5 + 6 antibody (ab17133) was ordered quite a long time ago. Almost a year prior to the report of the complaint. We would therefore not usually offer a replacement antibody as this requires that the complaint be made within 6 months of the purchase of the antibody to comply with the Abpromise guarantee. In this case I will offer a replacement vial of the antibody (ab17133) if your customer would like as a goodwill gesture.

I would however suggest that a few protocol alterations may be worthwhile in order to try to optimise the results observed.

1. I would suggest improving the specificity and strength of the detection by incubating with the primary antibody overnight as 4°C, not 37°C as is currently used. This antibody may not be suitable for use at elevated temperatures and we often find superior performance when used at lower temperatures for longer. This has been employed by the customer who has used this antibody previously and achieved the following staining:

https://www.abcam.com/index.html?datasheet=17133&tab=abreviews&intabreviewid=13948

2. You have mentioned that the antigen retireval has been optimised. How has this been performed? For this particular target or tissue? I would suggest trying the Citrate buffer currently used as well as Tris-EDTA (pH 9) under a few different times (3, 5, 10 and 20 minutes). This can often provide drastic differences in performance and optimisation may be required for each target you are interested in.

If you would like for me to add ab17133 to your next order please do let me know.

I look forward to hearing from you.

Thank you for taking the time to complete our questionnaire and contact us. I am sorry to hear you have had difficulty obtaining satisfactory results from this antibody.

The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality.

I would like to reassure you that ab17133 is tested and covered by our 6 month guarantee for use in IHC-P and human samples. In the event that a product is not functioning in the applications cited on the product data sheet, we will be pleased to provide a credit note or free of charge replacement.

Reviewing this case, I would appreciate if you can confirm some further details which will help me to investigate this case:

1. Please confirm the Abcam order reference number and date of purchase. According to our records, the last vial of this antibody purchased from ### was 14 months ago last March, order #####.

2. Which species is the sample from, is this human?

3. I can suggest to consider trying different times for antigen retreival. Has this already been tried? This can sometimes require some optimization.
