  • Description
Mouse monoclonal [Ks9.70/Ks9.216] to Cytokeratin 9
  • Host species
  • Specificity
Specifically reactive in the middle/upper suprabasal layers (stratum spinosum/granulosum) of the epidermis of palm and sole. CK9 can be detected in primary cultures of palmoplantar keratinocytes when they shift to differentiation-promoting conditions and grow stratified (upper cells). CK9 has not been found in epithelial cells of other stratified epithelia such as oesophagus or complex epithelia (e.g. urothelium) or in ductal or simple epithelia. Negative tissues include: muscle, liver and duodenum.

  • Species reactivity
    Reacts with: Human
  • Immunogen

    Synthetic peptide:

, corresponding to amino acids 450 - 477 of Human Cytokeratin 9. Synthetic peptide: RQFSSSYLTS GGGGGGGLGS GGSIR, corresponding to amino acids 4 - 28 of Human Cytokeratin 9.

Our Abpromise guarantee covers the use of ab19124 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use at an assay dependent concentration. Predicted molecular weight: 62 kDa.
IHC-Fr 1/20.
ICC/IF 1/10.
IHC-P 1/20. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.

Perform heat mediated antigen retrieval (microwave method) before commencing with IHC staining protocol; in some cases protease digestion is most suitable.

  • Function
May serve an important special function either in the mature palmar and plantar skin tissue or in the morphogenetic program of the formation of these tissues. Plays a role in keratin filament assembly.
  • Tissue specificity
Expressed in the terminally differentiated epidermis of palms and soles.
  • Involvement in disease
Defects in KRT9 are a cause of palmoplantar keratoderma epidermolytic (EPPK) [MIM:144200]; also abbreviated as EHPPK. EPPK is a dermatological disorder characterized by diffuse thickening of the epidermis on the entire surface of palms and soles sharply bordered with erythematous margins. Some patients may present with knuckle pads, thick pads of skin appearing over the proximal phalangeal joints.
  • Sequence similarities
Belongs to the intermediate filament family.

Thank you very much for your message which has been forwarded to me as my colleague Saun is no longer in the office. The blast results you have provided from rat, this indicates approximately 71% alignment. We recommend that alignment should be over 85% to predict that an antibody will detect in a different species. At 71%, this is quite low but there may be some crossreactivity. Therefore, I can offer a discount off a future purchase if you buy ab19124 now, test it in rat and submit feedback to us in the form of an Abreview. I have placed more details below. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback. The discount would be to the value of 1 free primary antibody. If you are interested in this offer, please follow these steps: 1. Reply to this e-mail to let me know that you would like to proceed and test ab19124 in rat. I will then send a discount code. This code must be issued before purchasing ab19124 so please wait for my reply before ordering. 2. Purchase ab19124 either by phone, fax, or online (www.abcam.com). 3. Test it in rat. 4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: https://www.abcam.com/abreviews. 5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any primary antibody ordered and the discount code is valid for 4 months after issue. We are always pleased to obtain feedback about our products and any information is greatly appreciated! Even if ab19124 turns out to be unsuitable for rat, you will still receive the discount on your next purchase after your Abreview has been submitted. I'm sorry to hear you are experiencing problems with ab19124. I would be grateful if you could please clarify a few details of your protocol so I can better understand your problem: could you confirm if you see the cytokeratin 9 band as well as a non specific band; could you also confirm the size of the non specific band you observe. It would be most useful to be able to see an image of your blot please. I would like to already suggest to decrease the blocking to 5% milk for 1 hour and to try to incubate the primary antibody in TBST only, as well as the secondary too. I would also recommend to try the secondary antibody alone to see if it is responsible for the non specific band. I look forward to hearing from you regarding details of the bands and hope the change in blocking will already help you.
