      This tyrosine kinase receptor for fibrillar collagen mediates fibroblast migration and proliferation. Contributes to cutaneous wound healing.
    • Tissue specificity

      The major 10 kDa transcript is expressed in high levels in heart and lung, less in brain, placenta, liver, skeletal muscle, pancreas, and kidney.
    • Involvement in disease

      Defects in DDR2 are the cause of spondyloepimetaphyseal dysplasia short limb-hand type (SEMD-SL) [MIM:271665]. A bone disease characterized by short-limbed dwarfism, a narrow chest with pectus excavatum, brachydactyly in the hands and feet, a characteristic craniofacial appearance and premature calcifications. The radiological findings are distinctive and comprise short long bones throughout the skeleton with striking epiphyses that are stippled, flattened and fragmented and flared, irregular metaphyses. Platyspondyly in the spine with wide intervertebral spaces is observed and some vertebral bodies are pear-shaped with central humps, anterior protrusions and posterior scalloping.

    Reviewing this case, I would like to offer some suggestions to help optimize the results. I would also appreciateif youare able toconfirm some further details:

    1. Please provide the order number and date of purchase.

    2. I am sorry to confirm that ab126773DDR2 antibody [EPR5179(2)] has not been tested in flow cytometry. Therefore, regrettably we are not able to provide the 6 month guarantee for this application.

    I can recommend to consider trying the antibody in WB, which has been tested and will be covered by the guarantee.

    2. I would appreciate if you are able to provide a copy of the histogram, which would help me to assess the results. Please confirm how the cells have been gated for the data, and a copy of the FS:SS plot would also be helpful.

    3. I can suggest to ensure the positive single color control is set up correctly on flow cytometer and gated/compensated correctly to capture all the events.

    5. For flow cytometry, antibodies are often required at very high concentrations, up to 1:10. I can recommend to increase concentration of antibody to help increase the signal.

    6. To prevent internalization of cell surface proteins, I would suggest all stagesof the staining procedurebe done on ice or at 4°C, with ice cold reagents, to stop all reactions. Adding sodium azide will prevent the modulation and internalization of surface antigens which can produce a loss of fluorescence intensity. For staining of cell lines, trypsin can often induce internalization of cell surface proteins, particularly cell surface molecules and more gentle detachment methods may be required, such as gentle scraping.

    7. We recommend cell lines should be 95% viable (by trypan blue staining) when harvested for staining. I can suggest to try a viability count.

    8. Have flow check beads and alignment been done?

    9. I can recommend to check the offset and gain. Use the positive control to set up the flow cytometer, using the offset to ensure the fluorescent signal from cells is not being cut off, and increase the gain to increase the signal (within reason – care should be taken).

    10. I can recommend to fix the cells with 0.1% PFA at the end of the staining, store the stained samples in the fridge at 4oC and read the results within 24 hours.
