Our Abpromise guarantee covers the use of ab61153 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
|ICC/IF||Use a concentration of 1 µg/ml.|
|IHC-P||1/50 - 1/100.|
|WB||1/500 - 1/1000. Detects a band of approximately 73 kDa (predicted molecular weight: 73 kDa).|
FunctionATP-dependent RNA helicase. Acts as a cofactor for XPO1-mediated nuclear export of incompletely spliced HIV-1 Rev RNAs. Also involved in HIV-1 replication. Interacts specifically with hepatitis C virus core protein resulting in a change in intracellular location.
Sequence similaritiesBelongs to the DEAD box helicase family. DDX3/DED1 subfamily.
Contains 1 helicase ATP-binding domain.
Contains 1 helicase C-terminal domain.
Cellular localizationNucleus speckle. Cytoplasm. Located predominantly in nuclear speckles and, at low levels, throughout the cytoplasm. Located to the outer side of nuclear pore complexes (NPC). Shuttles between the nucleus and the cytoplasm in a XPO1-dependent manner.

ab61153 at 1/50 dilution staining human brain tissue without (left) and with (right) blocking peptide; paraffin embedded.
ICC/IF image of ab61153 stained HepG2 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab61153, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
