Abcam is leading the way with our range of recombinant antibodies, knockout-validated antibodies and knockout cell lines, all of which support improved reproducibility.
We are also planning to innovate the way in which we present recommended applications and species on our product datasheets, so that only applications & species that have been tested in our own labs, our suppliers or by selected trusted collaborators are covered by our Abpromise™ guarantee.
In preparation for this, we have started to update the applications & species that this product is Abpromise guaranteed for.
We are also updating the applications & species that this product has been “predicted to work with,” however this information is not covered by our Abpromise guarantee.
Applications & species from publications and Abreviews that have not been tested in our own labs or in those of our suppliers are not covered by the Abpromise guarantee.

Our Abpromise guarantee covers the use of ab245429 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
|IP||Use at 2-5 µg/mg of lysate.|
|IHC-P||1/500 - 1/2000. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.|
FunctionInvolved in chromatin organization.
Tissue specificityUbiquitous. Expressed at relatively high levels.
Involvement in diseaseA chromosomal aberration involving DEK is found in a subset of acute myeloid leukemia (AML); also known as acute non-lymphocytic leukemia (PubMed:1549122). Translocation t(6;9)(p23;q34) with NUP214/CAN (PubMed:1549122). It results in the formation of a DEK-NUP214 fusion gene (PubMed:1549122).
Sequence similaritiesContains 1 SAP domain.
modificationsPhosphorylated by CK2. Phosphorylation fluctuates during the cell cycle with a moderate peak during G(1) phase, and weakens the binding of DEK to DNA.
Cellular localizationNucleus. Enriched in regions where chromatin is decondensed or sparse in the interphase nuclei.

Formalin-fixed, paraffin-embedded human prostate carcinoma tissue stained for DEK with ab245429 at a 1/1000 dilution in immunohistochemical analysis. DAB staining.
DEK was immunoprecipitated from HeLa (Human epithelial cell line from cervix adenocarcinoma) whole cell lysate (1 mg per IP reaction; 20% of IP loaded).
ab245429 used for IP at 3 µg/mg lysate. For WB 1 µg/ml.
