Question

As we have to prepare the sample 1: 50 dilution (20 ul of serum + 980ul of 1X Serum diluent) before add onto the plate wells, so for the final results, should we multiple it ?

For example : sample result was 1.365 ug/ml, and the normal range of adults is 0.9-3.6 ug/ml (by abcam)

If we multiple it, 1.365 x 50 = 68.25 ug/ml, it did not make sense.

Answer

The standard range of 0.1 - 10 ug/ml takes into account the 1/50 sample dilution that is done, so you do not need to multiply your end measurement by 50.
