    Could be involved in oogenesis. Involved in the regulation of endosome dynamics. Implicated in a novel signal transduction pathway, in which isoform 3 and CSK are sequentially activated by RHOD to regulate the motility of early endosomes through interactions with the actin cytoskeleton.

    Defects in DIAPH2 are the cause of premature ovarian failure type 2A (POF2A) [MIM:300511]. An ovarian disorder defined as the cessation of ovarian function under the age of 40 years. It is characterized by oligomenorrhea or amenorrhea, in the presence of elevated levels of serum gonadotropins and low estradiol.

  • IHC image of ab12319 staining in human normal testis formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab12319, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Answer

Thank you for your patience in this matter. I have received a reply from the originator of ab12319 (DIAPH2 antibody). The immunogenic peptide sequence is proprietary information. However, they indicated that this antibody recognizes an epitope that maps to a region between residues 50 and 100 of human Diaphanous Homolog 2 using the numbering given in entry NP_006720.1 (GeneID 1730). The crossreactivity of ab12319 with the other isoforms has not been specifically tested. I hope this information helps.
