Relevance
The three human RAS genes encode highly related 188 to 189 amino acid proteins, designated H-Ras, N-Ras and K-Ras4A and K-Ras4B (the two K-Ras proteins arise from alternative gene splicing). Ras proteins function as binary molecular switches that control intracellular signaling networks. Ras-regulated signal pathways control such processes as actin cytoskeletal integrity, proliferation, differentiation, cell adhesion, apoptosis, and cell migration. Ras and ras-related proteins are often deregulated in cancers, leading to increased invasion and metastasis, and decreased apoptosis. DIRAS1 displays low GTPase activity and exists predominantly in the GTP-bound form. It is highly expressed in heart and brain. -

Paraffin-embedded human brain tissue stained for DIRAS1 using ab238491 at 1/200 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at room temperature. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

A549 (Human lung carcinoma cell line) cells stained for DIRAS1 (green) using ab238491 at 1/66 dilution in ICC/IF. Secondary antibody is Alexa Fluor® 488-conjugated Goat Anti-Rabbit IgG (H+L).
The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the antibody overnight at 4°C.
