IHC-P Use a concentration of 10 µg/ml. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
Steam slides in 0.01 M sodium citrate buffer, pH 6.0 at 99-100°C for 20 minutes. Remove from heat and let stand at room temperature in buffer for 20 minutes. Rinse in 1X TBS with Tween (TBST) for 1 minute at RT.

    Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease.

  • ab61034 staining DKK1 in mouse embryonic (E16.5) craniofacial tissue by IHC-Fr (paraformaldehyde-fixed frozen sections).  Paraformaldehyde fixed issue samples were permealized in 0.1% Triton X-100, blocked with 5% serum for 15 minutes at 25 °C. The sample was incubated with primary antibody dilution 1/100 in PBS at 4°C for 24 hours. An Alexa Fluor®  conjugated Goat polyclonal to rabbit IgG (dilution 1/200)  was used as secondary antibody.

The details provided will enable us to investigate this case and will provide us with vital information for monitoring product quality. Having reviewed this case, I would like to offer some suggestions to help optimize the results from ab61034. Your customer may want to try a different blocking buffer than the one provided by Leica. I'm concerned that the blocking is interfering with the primary antibody binding. Try 5% BSA instead for 45 mins at RT. Also how long are they incubating the primary antibody? Because they are seeing such a low signal, it may be best to try incubating overnight at 4C. I can see that they tried our recommended 20 mins for antigen retrieval, but could they increase this to 25 or even 30 mins? If these suggestions do not work to improve your customer's results, please let us know and we would be happy to replace or refund the product for you.
