Target

  • Function

    Acts as a ligand for Notch receptors. Blocks the differentiation of progenitor cells into the B-cell lineage while promoting the emergence of a population of cells with the characteristics of a T-cell/NK-cell precursor.

      ab10554 staining DLL1 in U2OS cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with 4% paraformaldehyde, permeabilized with 0.3% Triton X-100 in PBS and blocked with 4% BSA for 1 hour. Samples were incubated with primary antibody for 1 hour at room temperature.       ab10554 staining DLL1 in Mouse embryonic kidney tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Samples were incubated with primary antibody (2ug/ml) for 1.5 hours at room temperature. A peroxidase-conjugated polyclonal was used as the secondary antibody. Nuclei stained purple with hematoxylin.

      ICC/IF image of ab10554 stained MCF7 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab10554, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

      Western blot using Delta 1 antibody (ab10554) at 1/500 tested on the following tissue lysates:

      Lane 6: Rat Brain

      Secondary ab: Goat anti-rabbit IgG ab6721 (1/5000)
      Exposure time: 1 minute
      Expected molecular weight: 77kDa
      Cell lysates at 20µg per lane.

      NB: This image shows that is was not possible to detect Delta 1 immunoreactivity by WB in the lysates tested. However little is known about Delta 1 expression and it is likely to be developmentally regulated with low expression/abundance making it difficult to detect in whole tissue homogenates. In favour of Delta 1 antibody (ab10554), the staining on the WB shows very low background and little cross reactivity in whole homogenates.

    Thank you for your email. Ab10554 is a Fast-Track antibody - Fast-Track antibodies are immunogen affinity purified and are shown by ELISA to have high titre values against their immunogen. We are currently testing them for use in Western blotting. At this stage, there can be delays. We sometimes experience problems in searching for / developing the correct lysate or knock-out cell-line. In addition, certain products can be difficult to characterise, due, for instance, to low expression levels or a lack of expression in easily available lysates. The preliminary characterization of this antibody has shown the following: by Western blotting (please see image on the online datasheet) it was not possible to detect Delta 1 immunoreactivity by WB in the rat and mouse lysates tested (brain, pancreas and heart). However little is known about Delta 1 expression and it is likely to be developmentally regulated with low expression/abundance making it difficult to detect in whole tissue homogenates. Mouse spleen has not been tested to our knowledge with this antibody. Please note that Fast-Track antibodies are released early with the following understanding: As Fast-Track antibodies are not yet fully characterised we are unable to offer our normal level of technical support. We cannot guarantee that the antibody will work in any application other than ELISA against the immunizing peptide and therefore we are unable to offer a refund if the antibody does not work in your application. However, Fast-Track antibodies are offered at a reduced price. We would greatly appreciate your feedback on these antibodies, whether positive or negative.     This Rabbit polyclonal antibody to Delta 1 has been shown to work in ELISA against the peptide immunogen but has not been shown to detect the endogenous protein (WB) in the lysates specified on the datsheet. The anitbody is sold as a special investigate grade called "Fast Track" and is offered at a reduced price (EUR 215 versus EUR 330). We explain the terms and conditions associated with fast-track antibodies on our datasheet. Furthermore we offer a sustantial number of reward points (currently 1000) to researchers who get back to us with positive or negative information about the antibody. So, there is no guarantee that this antibody will work in IHC and we do not offer free trial sized samples, but we believe that it is a sufficiently novel antibody to offer to the research community under the terms described above and in further detail on the datasheet.

    Thank you for your enquiry. We cannot give you an exact time before which the signal will fade with these fluorophore conjugated secondary antibodies. The signal could last a week (the time frame that you mentioned that you would need) if your samples are prepared and stored under the correct conditions (at 4C in the dark). However, we do not know this for sure. Also, some fluorophores will fade faster than others. Please note that ab10554 has not yet been tested for application in confocal microscopy.
