  • Function
    Methylates CpG residues. Preferentially methylates hemimethylated DNA. Associates with DNA replication sites in S phase maintaining the methylation pattern in the newly synthesized strand, that is essential for epigenetic inheritance. Associates with chromatin during G2 and M phases to maintain DNA methylation independently of replication. It is responsible for maintaining methylation patterns established in development. DNA methylation is coordinated with methylation of histones. Mediates transcriptional repression by direct binding to HDAC2. In association with DNMT3B and via the recruitment of CTCFL/BORIS, involved in activation of BAG1 gene expression by modulating dimethylation of promoter histone H3 at H3K4 and H3K9.

    Ubiquitous; highly expressed in fetal tissues, heart, kidney, placenta, peripheral blood mononuclear cells, and expressed at lower levels in spleen, lung, brain, small intestine, colon, liver, and skeletal muscle. Isoform 2 is less expressed than isoform 1.

I am not sure what the lower bands might represent. They are a bit low to represent any of the isoforms of DNMT1 in rat. A BLAST of the immunogen sequence did not reveal any homology with another protein that may be detected by this antibody. It would be interesting to see how the results compare to human samples (positive control). We have not specifically tested the antibody in rat and thus we have not performed any further analysis with this sample type.

I am sorry this product did not perform in rat as you had hoped and for the inconvenience this has caused. Since this antibody has not been tested in rat, we do not know if and how well it might work in this sample.

Unfortunately we do not have an antibody that has been specifically validated for rat, but I would like to recommend ab109981 as a potential alternative. This antibody is also predicted to work in rat and may be more suitable for your experiments.

I have been struggling with your Dnmt1 antibodies. 1. Ab2849 seems to work well for immunofluorescence (in the bovine) but gives me about 10 spare bands on the western human and bovine(1:1000 incubation 4C ON and blocking 5% skim milk). Those bands are not due to binding of the secondary antibody since I`ve tested it and different concentrations of the primary doesnt seem to help getting rid of those bands. The biggest problem is that the antibody binds the spare bands at a higher intensity than it does the band with the right predicted size. Did you ever characterize/sequence the bands you identified by western? I ran 2D transfers (westerns) using cell extracts of mcf-7 (human mammarian tumor cell line) and was unable to detect a spot at the right size and PI (180-190kDa and pI 7.4-8.0) instead I detected high molecular weight bands at pI 4-6 which are not Dnmts. 2. Ab4870 seems to be more specific (fewer spare bands) however it is very very weak! It does not work well on westerns. Both antibodies were used for immunoprecipitation and did not give me a band at the right predicted size. 3. Ab 14290 - you claim it works in the bovine. How has it been tested? Do you have protein sequence data proving it? If you do would you be able to send us a sample of it or trade a vial of the Ab4870 for one of the Ab1420. As you can see we have spent a lot of money and time buying and testing your antibodies so I would appreciate if we could get some technical support. Last time I contacted your company I was not able to get much information. All I got was a general protocol for westerns.

All the information that we have regarding these antibodies is located on the online datasheets; as these three antibodies were originated outside Abcam, I'm currently trying to obtain some more information from the originators to help you out. What were the lot numbers (located on the vials) that you received? What was the Abcam order number or purchase order number that was used? For ab4870, what were the dilutions that you tried and what type of samples did you use? Neither ab2849 or ab4870 have been tested before in IP, so there is no information regarding the use of the antibodies in that particular application.

The immunogen is a synthetic peptide conjugated to KLH, corresponding to within the last 100 amino acids of Human Dnmt 1. From analysing this sequence using Swiss Prot and BLAST I can see there is a 97% similarity between the human sequence and bovine.
