The attempt to detect localization of DNMT3b in a Rat hepatoma cell line by this antibody isn’t successful. Anti-RNA Pol II is used as positive control and normal rabbit IgG is used as negative control. Blast show in the region antibody raised rat and mouse have 100% identity. I used it for IF from one year and two years I ordered it, but I aliquoted and stored it very well and some others from the same purchase works. It is possible that the amount of DNMT3b in the cell nucleolus I detected is very low. It is also possible that conditions can be further optimized because of the same sequence between rat and mouse.

The results of your ICC experiment using rat hepatoma cells are certainly of interest as this application had not yet been tested for this antibody. ab2851 may not be suitable for immunocytochemistry. However, I agree with your assessment that, with optimization, this antibody may yet prove useful for this application.
