    By immunoelectrophoresis and ELISA this antibody reacts specifically with mouse IgG and with light chains common to other mouse immunoglobulins. No antibody was detected against non-immunoglobulin serum proteins. Less than 1% cross reactivity to bovine, chicken, goat, human, rabbit, rat and sheep IgG was detected. This antibody may cross react with IgG from other species.

  • Overlay histogram showing Jurkat cells stained with ab8090 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal donkey serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab8090, 0.01μg/1x106 cells) for 30 min at 22°C. The secondary antibody Donkey anti-mouse IgG H&L (Alexa Fluor® 488, pre-adsorbed) (ab150109) was used at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 0.01μg/1x106 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.

  • ICC/IF image of ab7291 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab7291, 10µg/ml) overnight at +4°C. The secondary antibody (green) was ab150109 Alexa Fluor® 488 donkey anti-mouse IgG (H+L) used at 2µg/ml for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Immunohistochemistry (PFA fixed)
Rat brain was perfusion-fixed using 4% PFA and cryo-sectioned in the coronal plane (20 µm thick).
Primary antibody: Mouse anti-NeuN antibody (Chemicon / Merck Millipore)
Secondary antibody: Donkey anti-mouse IgG H&L (Alexa Fluor® 488) preadsorbed (Abcam ab150109)
Blocking buffer: 10% Donkey serum, 3% BSA, 0.3% Triton X-100 in PBS (pH 7.4)
Brain section was blocked for 2 h at room temperature.
Incubation in primary antibody diluted in blocking buffer (1:400 dilution) at 4 °C for 16-18 h.
Incubation in secondary antibody diluted in PBS (1:2000 dilution) at room temperature for 2 h.
Image (scale bar = 200 µm) shown is taken from the cortical layer of the rat forebrain.
Strong fluorescent signal with moderate background noise can be observed.

We have not used this product with a LICOR system. Generally these systems use IR or fluorescent molecules such as Alexa Fluor 680, Alexa Fluor 750 for detection. We do not know if the system which you have will be able to excite and read the emission for an Alexa Fluor 488 fluorochrome. We suggest contacting your LICOR representative to determine if the system which you have will be able to do so.
