    ab7817 staining alpha smooth muscle actin in mouse primary colon myofibroblasts by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with acetone and blocked with 5% BSA for 30 minutes at 25°C. Samples were incubated with primary antibody (1/100 in PBS + 5% BSA) for 2 hour at 25°C. A DyLight® 488-conjugated donkey anti-mouse IgG H&L (ab96875) (1/1000) was used as the secondary antibody. Vimentin appears in red.

Dear xxxxxx,

Below is the reply from my customer:

I wish to use the antibodies in a co-localization experiment. We have primary antibodies against specific Plasmodium falciparum proteins which localize to two different parasite organelles. The fluorescent secondary antibodies will then allow these organelles to be visualized.

We have not performed an experiment like this in the lab before and we will have to develop a protocol. This is the reason why I was wondering what the recommended protocol would be for the DyLight secondary antibodies.

I'm on a very strict budget and I'm looking for the most cost effective way to perform this experiment. In comparison, the Alexa Fluor secondary antibodies have a working concentration of 1ug/ml, with a supplied concentration of 2mg/ml. Do the DyLight secondary antibodies, with a supplied concentration of 0.5mg/ml, have a lower working concentration than that of the Alexa Fluor?

Thank you for explaining this for me.

The antibodies mentioned, ab96875,ab96871,ab96877 andab96873 are all recommended to be used in a range of 1/50-1/500 for ICC and IF applications. This is only a starting point and optimisation of these conditions will be required by the end used and their particular experimental conditions. This will be the case with any secondary antibody. This equates to a final concentration of 1 to 10 ug/mL.

How many secondary antibodies are you looking to purchase? It may be that I can arrange for a discount for you?

May I know what kind of secondary antibody with FITC could be suitable for ab98952, for example, isGoat anti-Mouse IgG1 heavy chain (FITC) secondary antibody (a b97239) suitable? And I also wanna to know is there any conjugated antibody (conjugated withfluorescent secondary antibody) for N-cadherin? How about productAnti-N Cadherin antibody [GC-4], please recommend some for me, thanks a lot.
