    Answer

I’m sorry for the delay in getting back to you. I have discussed the problem the customer had been encountering with my colleague and as I mentioned the results they have been observing are what would be expected. The Alexa488 is a much stronger fluorophore than the PE conjugate and would therefore be expected to give a stronger signal. The ab7003 antibody being pre-adsorbed, whilst making it more specific for the particular species of antibody you are hoping to target, will also contribute to it being slightly less signal.
However, if your customer would like, I can offer for her to try an alternative antibody from our catalogue which may prove to give better results. I would suggest one of the following:
Ab99605 this is a rat monoclonal antibody and whilst it is still PE conjugated, it has been specifically enhanced to work in flow applications.
Alternatively, I can suggest altering the fluorochrome used to increase the signal such as using DyLight 650 or Cy5 (ab96882 or ab97037 respectively) if the customer has access to the appropriate lasers for excitation.
I would be willing to send the customer any of the suggested antibodies to try, or an alternative they may prefer. In order to arrange this could you please let me know which antibody they would like, as well as the original order number.
We are closed Monday and Tuesday of next week due the the Queens jubilee celebrations, I am sorry for any inconvenience this may cause.
I look forward to receiving your reply.

    Answer

Thank you for contacting us and sorry for the delay in getting back to you.
I have looked at the protocol your customer has been using and I would say that the results they have been observing are what would be expected.
I am going to speak to one of my collegues who is an expert in this area to find out if we might have a more suitable antibody for the experiment your customer is hoping to perform and will get back to you as soon as I have this information.
In the meantime I wish you a lovely weekend.

    Question

I would like to ask, in order to be absolutely certain, whether your secondary antibodies, mentioned below, contain Fc part: Goat F(ab')2 polyclonal Secondary Antibody to Mouse IgG - H&L (PE), pre-adsorbed (ab7002) Donkey F(ab')2 polyclonal Secondary Antibody to Mouse IgG - H&L (PE), pre-adsorbed (ab7003)   I would like to use them in the experiment where their binding to the Fc receptors would be an important issue.

    Answer

Thank you for contacting us.  Our F(ab')2 fragment secondary antibodies ab7002 and ab7003 will not contain the Fc region, and this will be covered under our Abpromise guarantee.  If any Fc reactivity is observed in your experiments, we would be happy to offer you a free of charge replacement, credit, or refund.  I hope this helps, please let me know if you need any additional information.
