  • Immunohistochemical analysis of paraffin-embedded human breast cancer tissue labeling DPEP3 with ab235451 at 1/4000 dilution, followed by Goat Anti-Rabbit IgG H&L (HRP) ready to use. Membranous staining on tumor epithelial cells of one case human breast cancer is observed. Counter stained with hematoxylin.

Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (HRP) ready to use.

Heat mediated antigen retrieval using ab93684 (Tris/EDTA buffer, pH 9.0).

Please note that DPEP3 was not detected in every breast cancer tissue sample tested due to the heterogeneity of expression.

  • Anti-DPEP3 antibody [EPR21862] (ab235451) at 1/1000 dilution + Human testis lysate at 20 µg

    Secondary
    Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/100000 dilution

    Predicted band size: 54 kDa
    Observed band size: 75 kDa
why is the actual band size different from the predicted?

    Exposure time: 48 seconds

Blocking and dilution buffer: 5% NFDM/TBST.

  • DPEP3 was immunoprecipitated from 0.35 mg of human testis lysate with ab235451 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab235451 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/5000 dilution.

Lane 1: Human testis lysate 10 μg (Input).
Lane 2: ab235451 IP in human testis lysate.
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab235451 in human testis lysate.

Blocking and dilution buffer and concentration: 5% NFDM/TBST.
    Exposure time: 3 minutes.

  • Immunohistochemical analysis of paraffin-embedded human testis tissue labeling DPEP3 with ab235451 at 1/4000 dilution, followed by Goat Anti-Rabbit IgG H&L (HRP) ready to use. Membranous and cytoplasmic staining on human germ cells but not Leydig cells (arrow) or Sertoli cells (arrowhead) (PMID: 21724266) is observed. Counter stained with hematoxylin.

Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (HRP) ready to use.

Heat mediated antigen retrieval using ab93684 (Tris/EDTA buffer, pH 9.0).
