    Defects in DRD5 are a cause of benign essential blepharospasm (BEB) [MIM:606798]. BEB is a primary focal dystonia affecting the orbicularis oculi muscles. Dystonia is defined by the presence of sustained involuntary muscle contraction, often leading to abnormal postures. BEB usually begins in middle age. Initial symptoms include eye irritation and frequent blinking, progressing to involuntary spasms of eyelid closure. Patients have normal eyes. The visual disturbance is due solely to the forced closure of the eyelids. In severe cases, this can lead to functional blindness.

  • ICC/IF image of ab40656 stained SKNSH cells. The cells were 4% formaldehyde (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab40656, 10µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

All our antibodies are for research purposes only and not for diagnostic or therapeutic ends. We therefore mainly test our products in in vitro application such as WB and IHC. All four antibodies mentioned have not been tested in vivo however this does not mean that they will not be suitable provided that this in vivo testing is not in humans and is in mouse or rat.

If you wanted to do in vivo testing, please note that Anti-Dopamine Transporter antibody (ab111468) and Anti-Dopamine Transporter antibody (ab111468) have 0.02% sodium azide in them as a preservative and that this would have to be removed before in vivo testing in rats as it could be harmful to them.

Cell harvesting/tissue preparation method: Spleen cell suspensions were obtained by gently grinding tissue into RPMI 1640 culture medium (Sigma-Aldrich, Steinheim, Germany) under sterile conditions. Mononuclear cells were separated by Ficoll–Hypaque density gradient (1.083 g/ml) centrifugation. The mononuclear cells concentrated at the surface were collected and washed twice in RPMI 1640 medium. After cell counting, 1 x 10 6 cells/sample were stained with the corresponding antibodies.
