    This antibody can only be used on highly purified dsDNA that has been stripped of chromatin. Histones can be removed by using an incubation step with proteinase K. We also recommend that you dilute no more than 1:10 - also try a 1:5 dilution.

    dsDNA (double stranded deoxyribonucleic acid) is the genetic material of all cells and many viruses and is a polymer of nucleotides. The monomer consists of phosphorylated 2-deoxyribose N-glycosidically linked to one of four bases, adenine, cytosine, guanine or thymine. These are linked together by 3',5'-phosphodiester bridges. In the Watson-Crick double-helix model, two complementary strands are wound in a right-handed helix and held together by hydrogen bonds between complementary base pairs.

This antibody has not been specifically tested against plasmids, however it should recognize double-stranded DNA free of chromatin as described on the datasheet. Thank you for your enquiry. I can tell you that the testing of this antibody was performed by immunohistochemistry using acetone fixed Hep2 and Hela cells. Unfortunately I do not have further details as to where this antibody has been applied on LR white resin embedded thin sections. The epitope of this antibody is dsDNA. Unfortunately I do not have specific information as to what within the DNA molecule it recognises other than that it recognises ds as opposed to ss DNA and requires a minimum of 10 bases (one complete helical turn) to recognise its epitope.
