Question

Would like to use one of these dynorphin A antibodies in formalin-fixed, free-floating tissue sections, but these antibodies recommend an antigen retrieval step. Is there a protocol that you recommend?

Answer

Thank you for your call yesterday and for your question about these antibodies. I apologize for the delay getting back to you.

We have a general IHC protocol for FFPE sections that includes a section on antigen retrieval-

https://www.abcam.com/index.html?pageconfig=resource&rid=11488

However, these methods are optimized for slide-mounted tissue, so I have found some other resources that would be more useful for free floation sections-

http://www.ihcworld.com/_protocols/epitope_retrieval/free_floating_sections.htm

I've also attached a couple references for free floating antigen retrieval. You should be able to perform this step in a 24-well plate so it will be compatible with your current staining protocol.

I hope this information will be useful, but if you have any further questions or need anything else, please let me know and I'll be happy to help you further.
