Target

  • Function

    Cadherins are calcium-dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types. CDH1 is involved in mechanisms regulating cell-cell adhesions, mobility and proliferation of epithelial cells. Has a potent invasive suppressor role. It is a ligand for integrin alpha-E/beta-7.
    E-Cad/CTF2 promotes non-amyloidogenic degradation of Abeta precursors. Has a strong inhibitory effect on APP C99 and C83 production.

  • Involvement in disease

    Defects in CDH1 are the cause of hereditary diffuse gastric cancer (HDGC) [MIM:137215]. An autosomal dominant cancer predisposition syndrome with increased susceptibility to diffuse gastric cancer. Diffuse gastric cancer is a malignant disease characterized by poorly differentiated infiltrating lesions resulting in thickening of the stomach. Malignant tumors start in the stomach, can spread to the esophagus or the small intestine, and can extend through the stomach wall to nearby lymph nodes and organs. It also can metastasize to other parts of the body. Note=Heterozygous germline mutations CDH1 are responsible for familial cases of diffuse gastric cancer. Somatic mutations in the has also been found in patients with sporadic diffuse gastric cancer and lobular breast cancer.
    Defects in CDH1 are a cause of susceptibility to endometrial cancer (ENDMC) [MIM:608089].
    Defects in CDH1 are a cause of susceptibility to ovarian cancer (OC) [MIM:167000]. Ovarian cancer common malignancy originating from ovarian tissue. Although many histologic types of ovarian neoplasms have been described, epithelial ovarian carcinoma is the most common form. Ovarian cancers are often asymptomatic and the recognized signs and symptoms, even of late-stage disease, are vague. Consequently, most patients are diagnosed with advanced disease.

    modifications

    During apoptosis or with calcium influx, cleaved by a membrane-bound metalloproteinase (ADAM10), PS1/gamma-secretase and caspase-3 to produce fragments of about 38 kDa (E-CAD/CTF1), 33 kDa (E-CAD/CTF2) and 29 kDa (E-CAD/CTF3), respectively. Processing by the metalloproteinase, induced by calcium influx, causes disruption of cell-cell adhesion and the subsequent release of beta-catenin into the cytoplasm. The residual membrane-tethered cleavage product is rapidly degraded via an intracellular proteolytic pathway. Cleavage by caspase-3 releases the cytoplasmic tail resulting in disintegration of the actin microfilament system. The gamma-secretase-mediated cleavage promotes disassembly of adherens junctions.

    Cell junction. Cell membrane. Endosome. Golgi apparatus > trans-Golgi network. Colocalizes with DLGAP5 at sites of cell-cell contact in intestinal epithelial cells. Anchored to actin microfilaments through association with alpha-, beta- and gamma-catenin. Sequential proteolysis induced by apoptosis or calcium influx, results in translocation from sites of cell-cell contact to the cytoplasm. Colocalizes with RAB11A endosomes during its transport from the Golgi apparatus to the plasma membrane.

      ab15148 staining E Cadherin in Pig Cervix uteri tissue sections by IHC-P (Formaldehyde-fixed, Paraffin-embedded sections). Tissue samples were fixed with formaldehyde and blocked with 10% goat serum for 1 hour at 37°C; antigen retrieval was by heat mediation in 10mM citrate at pH 6 for 2 minutes. The sample was incubated with primary antibody (1/50) at 4°C for 12 hours. An HRP-conjugated goat polyclonal to rabbit IgG (undiluted) was used as secondary antibody.

      ab15148 staining E Cadherin in human stomach tissue section by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue underwent formaldehyde fixation before heat mediated antigen retrieval in Citrate pH 6.0 and then blocking with 5% serum for 1 hour at 23°C was performed.     BATCH NUMBER -- NOT SPECIFIED -- ORDER NUMBER -- NOT SPECIFIED -- DESCRIPTION OF THE PROBLEM No staining or very weak staining. I have tried staining on paraffing embedded sections and frozen/OCT embedded sections wihout much success. SAMPLE E5-E10 of chicken gizzard epithelium PRIMARY ANTIBODY Abcam: Rabbit polyclonal to E-cadherin (ab15148). We have used this antibody at different dilutions. Apparently it is recommended to be used at 1:30 dilution but in addition to that dilution we have used 1:15, 1:10 and 1:5 with little success. The antibodies I usually leave overnight at 4 C. SECONDARY ANTIBODY Jackson ImmunoResearch Laboratories Inc. Texas Red conjugated AffiniPure Goat anti-Rabbit IgG (H+L). Code Number: 111-075-144. Concentration 1.5mg/mL. Stored at 4 C in dark. DETECTION METHOD Immmunofluorescence method (Texas Red conjugated) POSITIVE AND NEGATIVE CONTROLS USED none for now ANTIBODY STORAGE CONDITIONS Rabbit polyclonal to E-cadherin antibody has been kept at 4 C since it was recieved on Nov 3. FIXATION OF SAMPLE The embryos were fixed in 4% paraformaldehyde for 1 hr after dissection and additionally for 10 minutes after rehydrating the slides prior to adding primary antibodies. ANTIGEN RETRIEVAL I have tried both: citrate buffer method, pH 5.0 and proteinase K treatment for 20 minutes. Citrate buffer method worked better but at that point I got way too much of background noise even after I added the secondaries at 1:200 dilution. PERMEABILIZATION STEP We don't use TritonX-100. But we do use PBS + tween on all of our washes. BLOCKING CONDITIONS We use 10% goat serum in PBT for blocking solution at two steps. The first time we use the blocking solution is after refixing the slides with 4% PFA (as I explained above) and before adding primary antibody. Two, we use 1% goat serum in PBT after allowing primary antibodies on all night at 4 C. HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 6 HAVE YOU RUN A "NO PRIMARY" CONTROL? No DO YOU OBTAIN THE SAME RESULTS EVERY TIME? No WHAT STEPS HAVE YOU ALTERED? I have tried to change embedding media but with no success. ADDITIONAL NOTES Here is the full copy of my protocol: 1) Xylene 2X5minutes 2) 1 min 100% ETOH 3) 1 min 100% ETOH 4) 1 min 75 % ETOH 5) 1 min 50 % ETOH 6) 1 min 25 % ETOH 7) 5 min PBS 8) 5 min PBS 9) 10 min fresh ice cold 4 % paraformaldehyde 10) 2x5 min PBS 11) 10 % goat serum in PBT 12) Primary antibody (Dilute Abs in PBT, plus 1% serum: lay flat in humidifieed chamber, 400ul/slide. 4 C overnight) 13) 3x10 min 1% goat serum in PBT 14) Secondary Antibodies. 1 hr in dark, dilute at 1:200, add DAPI. Lay slides flat on humidified chamber. 400ul/slide. Wrap containier in foil. 15) 3X10 min PBT in dark 16) 1X10 min PBS in dark 17)Mount slides using VECTASHIELD and nail polish to seal dges. Store at 4 C in the dark.

    I'm sorry you are having problems with ab15148 on your chicken tissue. Thank you for taking the time to fill in the questionnaire. This antibody has not been tested in chicken. I have looked at the information and think there is no problem with your staining protocol. I have done a BLAST search of the chicken genome (gallus gallus) with the immunogen sequence IFFCERNPKP QVINIIDADL PPNTSPFTAE LTHGASANWT IQYNDPTQES IILKPKMALE VGDYKINLKL MDNQNKDQVT TLEVSVCDCE GAAGVCRKAQ PVEAGLQI , corresponding to amino acids 600-707 of Human E Cadherin and found very little identity (43%). Furthermore, there are no small sequences (5-6 amino acids long) identical, it is therefore very likely your staining is very weak in chicken tissue because the antibody does not recognise well chicken E-cadherin.
