Target

  • Relevance

Escherichia coli is a gram negative bacillus that belongs to a larger group of Enterobacteriae - bacteria that inhabit the gastrointestinal tract. Although usually a harmless resident of the gut, some strains have the potential to cause serious problems, especially where there is an immature immune system or immunosuppression, or where the subtype of organism has acquired the ability to produce pathogenic toxins.

Answer

Thank you so much for your patience while I have worked on this enquiry.

I've looked through the literature, data on our E.coli antibodies, and also discussed with my colleagues, and I think that the unconjugated version of ab20640 will be the best option for isolating E.coli as you describe. The immunogen was a mixture of mostly intact bacteria of various serotypes, and the rabbit IgG will have a higher affinity for Protein A compared to the goat antibodies, which haveonly a moderate affinity for protein G. I'm currently working to add this unconjugated antibody to our catalog, and I'll let you know as soon as this is done. I expect it will be donesoon.

Thank you again for your patience, and please let me know if you have any questions or need anything else in the meantime.

Answer

Thank you for your reply.

I will get in touch with the lab about the unconjugated E. coli antibodies that we have, and see if there is any data to suggest that one will work in your assay! I'll get back in touch with you as soon as possible.

Please let me know if you need anything else in the meantime.

Answer

Thank you very much for your reply and for sending the description of the assay in which you will be using the E. coli antibody.

We have not tested any of our E. coli antibodies in a similar assay, so there isn't one in particular that we know will work in this assay. I think that the unconjugated antibody used with Protein A/G beads would be the best protocol to try since it has worked with other antibodies and cells, however we do not have an E. coli antibody that was raised against whole intact cells like the S. cerevisiae antibody ab19498 was. As previously mentioned, if you would like to submit a review of ab68451 in this assay, then I can provide you with a discount code that can be redeemed for one free full size antibody. So, you will be able to try a different E. coli antibody at no charge. The goat polyclonal antibodies ab13627, ab48416,and ab25823 are the only unconjugated, polyclonal E. coli antibodies that we currently sell, so you may want to try one of these. When using a goat antibody, Protein G beads will need to be substituted for the Protein A beads.

If you would like to accept this offer, please let me know and I will set-up the discount code for you. Please let me know if you have any questions or need anything else at this time.

Question
Answer

Thank you for your reply.

I am very sorry that we do not have free samples to send, but our testing program is the option that weprovide for trying an antibody in an untested application.

We generally arrange this program prior to purchase of the antibody. However, if you would like to submit a review for ab68451 in the assay that you have performed, I can set the program up for you now. This way you can receive a different antibody to try for free. Please let me know if this is a suitable arrangement and I will send you the details.

I look forward to hearing from you. Please let me know if you have any questions, and I'll be happy to help.

Answer

Thank you very much for your reply.

At this time, we do not have the unconjugated format of ab68451 in our catalog, but if you are interested in trying it Iwill see if we can add this antibodyto ourcatalog. We also have two unconjugated goat polyclonal antibodies, ab13627 and ab25823, which may be suitable as well-

https://www.abcam.com/E-coli-antibody-ab13627.html

https://www.abcam.com/E-coli-antibody-ab25823.html

Protein G beads should be used with the goat antibodies, as goat IgG has little affinity to protein A.

We only have full size vials of all of our antibodies, so unfortunately we do not have free samples to send out. These antibodies have not been tested ina bacterial pulldown assay, but if you would like to try one of these other unconjugated E. coli antibodies, we do have a testing discount program that you may be interested in. As part of this program, you would initially purchase the unconjugated E. coli antibody, test it in your assay, then submit an Abreview with your data and protocol. We will then give you one free primary antibody on a future purchase. I can send more information about this program if you would like.

Please let me know if you are interested in any of these other E.coli antibodies or if you have further questions, and I'll be happy to help.

Answer

Thank you for your call last week and for your patience while I have been in touch with the lab regarding your enquiry.
I have confirmed with the lab that the biotinyalted antibody is tested, and I've obtained the following ELISA protocol from the lab:
a) Fill wells of microtiter plate with 100 ul of bacterial suspension that is just visibly “hazy” in PBS or equivalent buffer. Incubate overnight uncovered@ 37 C in order to dry down the suspensions in the wells. Leave some wells uncoated as controls.
b) Wash wells 3 times with PBS containing 0.3% tween-20 detergent (PBST), or equivalent.
c) block wells with detergent solution containing 1 mg/ml BSA (gelatin or powdered, non-fat milk may be substituted) for a minimum of 1 hour @ RT.
d) repeat step b
e) add 100 ul of dilutions (made with PBST) of known postive and negative sera (or antibody being tested) to the various wells and incubate for 30 min. @ RT.
f) repeat step b
g) add 100 ul of Streptavidin-HRP diluted optimumly in PBST to all the wells and incubate for 30 min. @ RT.
h) repeat step b
i) add 100 ul of enzyme substrate solution to all wells and monitor the color development (visually or instrumentally).
I hope that this protocol will be useful, but if there is anything else that we can do for you please let me know and I'll be happy to help.

Answer

Thank you for your enquiry. I am afraid that there is no information on which surface protein that this product detects. The originator of the product informed us that the immunogen for this ab68451 is a mixture of all antigenic serotypes of Escherichia coli. It reacts with all "O" and "K" antigenic serotypes of E. coli. Since this antibody is unabsorbed, it will likely cross-react with related Enterobacteriaceae. I sincerely apologize that we seem to had a datasheet error. This product has been successfully used in ELISA applications but unfortunately has yet to be tested in ICC/IF, IHC-Fr, and WB as previously stated on the datasheet. I have removed these applications from the datasheet but please rest assure that this does not mean that the product will not work in those applications, just that we have not tested them and will need to be ascertained by our laboratory or the end user. All of our products are guaranteed to work in species and application as stated on the datasheet. Should you do not get expected results, then please inform us within the guarantee period of 120 days. We will help troubleshoot the experiment and if the product is indeed faulty, we will be more than happy to offer a free replacement or refund. I hope our Abpromise guarantee will give you the assurance you need to use this product in your research.
