Application Abreviews Notes
Flow Cyt 1/50.

ab199091 - Rabbit monoclonal IgG (Alexa Fluor® 488), is suitable for use as an isotype control with this antibody.

  • Function

Binds phospholipid vesicles containing phosphatidylinositol 3-phosphate and participates in endosomal trafficking.
  • Sequence similarities

    Contains 1 C2H2-type zinc finger.
    Contains 1 FYVE-type zinc finger.
  • Domain

The FYVE-type zinc finger domain mediates interactions with phosphatidylinositol 3-phosphate in membranes of early endosomes and penetrates bilayers. The FYVE domain insertion into PtdIns(3)P-enriched membranes is substantially increased in acidic conditions.
  • Cellular localization

    Cytoplasm. Early endosome membrane.

  • Overlay histogram showing HeLa cells stained with ab185039 (red line). The cells were fixed with 4% formaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab185039, 1/50 dilution) for 30 min at 22ºC. Isotype control antibody (black line) was rabbit IgG (monoclonal) Alexa Fluor® 488 used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.

Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.

This antibody gave a positive signal in HeLa fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.
