Inquiry: Hi, I am struggling to detect a signal for recombinant hEGF (ADI-908-060-100) on a dot blot using this antibody. Would it be possible to get the protocol that was used for testing the antibody originally? I am using the snapID system from Millipore, and the parameters I have used are 5-40ng protein loaded, denatured with SDS and reduced with 5% beta mercaptoethanol and heated for 5 min at 95 degrees. blocking with either 0.5% skim milk or 1% BSA primary antibody 0.6ug/ml, secondary (31460, Pierce) at 1/60,000, both diluted in blocking buffer. Times and wash steps are per SnapID instructions. Detection is via ECL. Any help would be greatly appreciated,
