Human IgM ELISA Kit

Human IgM ELISA Kit (ab214568) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IgM protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, milk, saliva, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IgM with 50 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. CALIBRATION

ab214568 (IgM) - This immunoassy is calibrated against a highly purified human IgM. The NIBSC/WHO unclassified purified human IgG/IgM/IgG preperation 67/086. was evaluatied in this kit.

The dose response curve of the unclassified standard parallels the SimpleStep standard curve. To convert sample values obtained with the SimpleStep IgM kit to approximate NIBSC International units, use the equation below.

Immunoglobulin M, or IgM, is a basic antibody produced by B cells and the largest of the immunoglobulins in the Human circulatory system. IgM typically constitutes about 10% of serum immunoglobulins and is prominent in the early immune responses. IgM (with IgD) is the major immunoglobulin expressed on the surface of B cells. IgM typically forms pentamers which includes the J chain, while in the less common hexameric form the J chain is absent. Although the common pentameric form of IgM contains 10 antigen binding sites, IgM cannot bind 10 antigens simultaneously due to size constraints. Class specific anti-immunoglobulin antibodies are useful for the characterization of malignant B cell proliferations. Most lymphocytic leukemias share either surface or intra cytoplasmic Ig with an isotypic restriction, which suggest the monoclonal nature of the cell population. Most of the chronic lymphocytic leukemias, non Hodgkin lymphomas and Burkitt's lymphoma bear surface IgM, whereas plasmocytes from Waldenström's disease bear intracytoplasmic IgM.
