5 mg of lyophilized microdissected skeletal muscle (vastus laterals) was homogenized in 180 uL 1X cell lysis buffer (included in kit). After a 10,000 x g 5 min spin at 4°C removed the supernatant was removed and the total protein determined. 250 μg of total protein was added to each membrane with overnight incubations for each incubation step. The attached image is the first blot setting up the method and was exposed using a FluorChem M for 27 seconds. As can be seen Acrp30 will require less protein/less exposure but otherwise all but seven cytokines were visualized following this protocol.
